In this study,ISSR and SRAP molecular markers were used to analyze the genetic diversity of Hemsleya from Sichuan,Guizhou,Hubei,Yunnan,and Chongqing,and a method was established to simultaneously determine the Components of Cucurbitacin ?b,Chikusetsu saponin ?a and Cucurbitacin ?a by High Performance Liquid Chromatography.To provide scientific guidance and basis for the rational development and effective use of Hemsleya resources.The results were as follows:1.ISSR: a total of 241 bands were amplified from the selected 18 primers,of which 97.09% were polymorphic bands.The genetic similarity(Gs)coefficients among the 19 materials ranged from 0.652 to 0.851 with an average of 0.752.And the UPGMA cluster dendrogram clustered into 5 categories at a Gs of 0.754.SRAP: the primers with 10 pairs of upstream primers with 5' fluorescent tags as IRDye-700 were amplified to obtain 609 bands,of which 95.57% were polymorphic bands.The Gs between the 19 materials ranged from 0.646 to 0.857 with an average of 0.740.The dendrogram constructed by the UPGMA method clustered into three categories at a Gs of 0.730.2.The correlation coefficient between ISSR and SRAP is 0.8441.It shows that the two molecular markers have high consistency and reliability,which can be used to study the genetic diversity of Hemsleya.Based on the ISSR and SRAP data,the genetic similarity coefficients among 19 samples of Hemsleya were between 0.662 and 0.845,with an average of 0.744.The dendrogram constructed by the UPGMA method clustered into three categories at a Gs of 0.730.Based on the numerical analysis of RP,PPB and EMR,SRAP molecular markers are more suitable for studying the genetic diversity of Cyclobalanopsis.3.A method for the simultaneous determination of Cucurbitacin ?b,Chikusetsu saponin ?a and Cucurbitacin ?a in Hemsleya by HPLC was established.Quantitative analysis showed that the content of Cucurbitacin ?b in 11 species of Hemsleya was in the range of 0.479~10.234 mg/g,and the content in H.megathyrsa was the highest.The content of Chikusetsu saponin ?a is in the range of 1.210~18.58 mg/g,the content of H.omeiensis is the highest.Cucurbitacin ?a content in the range of 3.079~20.76 mg/g,the content of H.pengxianensis var.jinfushanensis is the highest.
