| Because of its own physiological characteristics,weaned piglets grow fast and are highly susceptible to diseases.Typhoid hardship will lead to their weak temper.Weaning and circle stress will reduce their immunity.The "care" of weaned piglets is "life-saving".According to its characteristics,the effects of "invigorating spleen and replenishing qi,nourishing kidney and benefiting essence" were studied on the growth performance and immune function of conservation pigs.In this study,through screening "Jianpi Yiqi,Bushen Yijing" Chinese herbal compound,optimizing the preparation method of granules,preliminary study on quality standards,study on the effect of "Shenqi" granules on growth performance and immune function of fattening pigs,to provide theoretical basis for promoting its application in pig production.Specific research contents are as follows:(1)18 traditional Chinese medicines were screened by immune function and growth performance,and the compound was composed of Codonopsis pilosula and Lycium barbarum.(2)Single factor and Box-Behnken response surface methodology were used to determine the optimum extraction times of Codonopsis pilosula,extraction time was 40 minutes,ratio of material to liquid was 1:47,the yield of Codonopsis pilosula polysaccharides was 23.67%,and that of Lycium barbarum was 4 times,50 minutes,ratio of material to liquid was 1:43,and the yield of Codonopsis pilosula polysaccharides was 9.37%.(3)The extracted liquid was concentrated to a density of 1.20-1.22 for wet granulation.Soluble starch was screened out by shaping rate.Ethanol volume fraction was fixed at 70%.The granule size qualification rate,solubility and moisture absorption rate were taken as the evaluation indexes.The optimum granule preparation process was wet granulation by mixing liquid with 70%soluble starch with density of about 1.20.(4)Preparing granules by optimum process and preliminary study on their quality.The appearance,particle size,water content and solubility of the granules were examined.The prepared Chinese medicine granules were yellow-brown with a moulding rate of 94.56%,water content of 4.22%,dissolution time of4.08 min,slightly turbid solution,angle of repose of about 30 degrees and critical relative humidity of granules of about 76%.(5)Single factor complete randomized trial design was used to select piglets weaned at 26 days old,injected with one dose of attenuated vaccine of classical swine fever at 30 days old,and blood samples were collected at 35 days old to detect the antibody level of classical swine fever.Then 90 "Du *Chang *Da" hybrid weaned piglets with similar clinical health and antibody level were selected and divided into five groups according to the principle of similar body weight.** Group Ⅰ was fed basal diet,and Ⅱ,Ⅲ and Ⅳ groups were added "Shenqi" granule 0.5%,1% and 2%(feed ratio to0.0625g/kg,0.125g/kg,0.25g/kg)respectively in feed.Ⅴ group added Astragalus Polysaccharides in feed,started at 35 days of age,piglets 63 days old,and ended with conservation.The experimental period was 28 days.Six piglets were randomly selected from each group at 35,42,49,56 and 63 days of age.Blood samples were collected from anterior vena cava for detection of IgG,IgA,IgM and CSF antibody levels.The proportion of feed to weight in group Ⅲ,Ⅳ and Ⅴ was significantly higher than that in control group(P < 0.05);the contents of IgG,IgA and IgM in serum of piglets in group Ⅱ,Ⅲ and Ⅳ were significantly increased(P < 0.05 or P < 0.01),and the effect was most obvious after feeding for more than two weeks;the level of antibody to classical swine fever in drug group was effectively increased(P < 0.05 or P < 0.01).The effect of continuous feeding with medium dose for more than 2 weeks was the most obvious.This study showed that the preparation process of the granule was scientific and reasonable,the quality standard was effective and controllable,and the screening and granulation conditions were feasible.It could significantly improve the antibody level and immunoglobulin level of the fever virus in weaned piglets,thereby improving the immunity of piglets.Adding 0.5% dosage to the diet for more than 2 weeks was the best. |
