Growth hormone(GH)can produce mass of biological actions on vertebrates including advancement of body growth,stimulation of arbitrator metabolism and transcription regulation of precise genes.These biological activities are facilitated through the growth hormone receptors(GHRs)on the target tissues.The target cells responsive to growth hormone contain the membrane-bound GHRs.Similar to other cytokine receptors;the signal transduction event is initiated by one GH binding to two GHRs to form a dimer of receptors.Similarly,Steroid hormones play essential roles in the generative biology of vertebrates.Several of the presently recounted activities of steroid hormones,comprising estrogens,androgens,and progestogens,are mediated by precise receptors that are confined in the nucleus of target cells.There are at least three estrogen receptors(ERs),ERa,ERβ1,and ERβ2,exist in teleost fish,which are ligand,activated transcription factors.Estrogen which exercises its effect through estrogen receptors is not solitary a feminine generative hormone but acts virtually all over in the vertebrates’ physique including fish and is involved in physiological and pathological states in all sexes of males and females.In human,for instance,estrogen has possessions in mandible conservation and the circulatory and central nervous systems as well as the generative system.There is relatively little knowledge of growth hormone,growth hormone receptors and estrogen receptors in Hybrid vertebrates especially Hybrid grouper.Therefore,this thesis aims to use Hybrid grouper(Epinephelus fuscoguttatus ♀×Epinephelus polyphekadion ♂)as models to study the GH,GHRs,and ERs in Hybrid vertebrates.The specific aims were to investigate if growth hormone,growth hormone receptors(GHR1&GHR2)and estrogen receptors(ERα,ERβ1&ERβ2)are present in the Hybrid grouper there after their expression was also studied in the Hybrid grouper.The Hybrid grouper GH,GHRs and ERs encoding cDNA were sequestered from the liver and heart cDNA library.Sequence analysis of GH showed the 5’untranslated region of 146 bp,open reading frames of 588bp,and 3’ untranslated region of 166bp,making the total length of 899 bp,and encoding protein with 156 amino acid residues.GHR1 showed 5’UTR of 6bp,open reading frames of 1842 bp,and 3’UTR of 328 bp,making the total length of 2176 bp,and encoding proteins with 612 amino acid residues.GHR2 was established to consist 5’UTR of 48 bp,open reading frames of 708 bp and 3’UTR of 1068 bp making the total length of 1824 bp encoding proteins with 235 amino acid residues.On the other hand,ERα showed 5’utr of 172 bp,open reading frames of 1866 bp,and 3’UTR of 354 bp,making the total length of 2391 bp,and encoding proteins with 621 amino acid residues.ERβ1 was established to consist 5’utr of 53 bp,open reading frames of 1638 bp and 3’UTR of 457bp making the total length of 2626 bp encoding proteins with 546 amino acid residues.ERβ2 was established to consist 5’UTR of 1075 bp,open reading frames of 732 bp and 3’UTR of 532 bp making the total length of 2339 bp encoding proteins with 244 amino acid residues.These results confirmed the identities of GH/GHRs and ERs in hybrid grouper which are full of biological structures and characteristics and open way for further studies for better ways to improve the growth rate of hybrid grouper.The findings provide vital statistics of growth hormone,growth hormone receptors and estrogen receptors likely to play a significant role in the growth of the fish. |
