MYB transcription factors play an important role in the growth and secondary metabolism of plants.However,MYB transcription factors in dandelion have not been reported.The Ta MYB59 transcription factor was cloned from Dandong dandelion in this laboratory,and the Ta MYB59 transcription factor was initially evaluated.The specific results of the test are as follows:1.The cloning and amplification of the Ta MYB59 transcription factor gene sequence,the open reading frame(ORF)size of 732 bp,encoding 243 protein genes,named Ta MYB59.This transcription factor has conserved domain R2 and R3 domains and is a member of the typical R2R3-MYB family of genes.2.The expression of Ta MYB59 gene was investigated by q RT-PCR.The results showed that the tissue-specific analysis showed that Ta MYB59 had the highest expression in the flower parts of Dandong dandelion.After salt stress and salicylic acid treatment,the expression of Ta MYB59 gene increased first and then decreased.After Na Cl treatment for 3 hours,it increased 22 times compared with the control group and 14 times after 3 hours after SA treatment.After treatment with methyl jasmonate,the relative expression of Ta MYB59 was not significantly different.3.The Ta MYB59 gene was analyzed by bioinformatics method,and the prokaryotic expression vector p Cold-Ta MYB59 was constructed.The size of Ta MYB59 recombinant protein obtained by SDS-PAGE was 40 Kd,which was consistent with the predicted size.Subcellular localization analysis of Ta MYB59 protein and construction of 35S-Ta MYB59-GFP expression vector showed that Ta MYB59 protein was localized in the nucleus.4.Analyze the ANS gene promoter containing 6 MBS cis-acting elements.The AD-Ta MYB59 and p Ab Ai-pro Ta ANS vectors were constructed,and the results of yeast one-hybrid showed that AD-Ta MYB59 could bind to the MBS cis-acting element.
