Plant regeneration was completed using stem segments as explants in Prunus mira Koehne.A series of experiments were carried out to investigate the effects of,basic media,time of explant isolation,explant sterilization,growth regulators on plant regeneration of stem segments sprouting,multiplication,rooting and transplantation.The results are as follows.(1)The results showed that an optimal disinfection result could be obtained in 75%alcohol for 20s+0.1%mercuric chloride for 13min,The best season for isolating explants was from March to April with the highest explant germination rate and lowest browning rate and contamination rate,the suitable basic medium for the bud induction was MS.(2)The appropriate initial culture medium was MS+6-BA0.5mg·L-1PPPP+NAA 0.2mg·L-1PPPP,in which the sprouting rate and average shoot number was 75.21%and 2cm,respectively.(3)The suitable shoot proliferation medium of the light walnut was MS+6-BA0.5mg·L-1PPPP+NAA0.3mg·L-1PPPP+CH 125mg·L-1PPPP,which resulted in multiplication coefficient of 4.91 and good plantlet growth status.6-BA,NAA and CH had extremely significant effect on shoot proliferation,and 6-BA played the biggest role in shoot proliferation.(4)The appropriate rooting medium of the light walnut was 1/2MS+NAA 0.25mg·L-1PPPP+IBA 1.0 mg·L-1PPPPwith the rooting rate of 88.05%,average root number was 16.59and average root length was 3.64cm with good plantlet growth status.(5)The rooted plantlets,after hardening,survived at a rate of 88.44%with well-developed and deep green leaves,thick shoots and good plant growth status in the transplantation matrix of decomposed garden soil,organic fertilizer(1:2)under careful temperature and water management. |
