In this experiment,the effects of different concentrations(5,10,20 mmol?L-1)of CaCl2 on the contents of endogenous hormones(ABA,GA3,IAA,ZT)in Sour jujube seedlings were studied under the stress of 150 mmol?L-11 NaCl.The Sour jujube seedlings were treated with150 mmol?L-11 NaCl and 150mmol?L-11 NaCl+10mmol?L-11 CaCl2 for 6 hours respectively.Then the transcriptome sequencing and assembly of the leaves and roots of the Sour jujube seedlings were carried out by high-throughput sequencing technology.All Unigene performs functional annotations in the GO and KEGG databases.To investigate the effect of exogenous CaCl2 on the content of endogenous hormones in Sour jujube under NaCl stress and transcriptome sequencing analysis,in order to further study the molecular mechanism of CaCl2 alleviating the salt damage of Sour jujube and provide reference for mining related salt-tolerant genes.The research results are as follows:1.Under NaCl stress,5,10,20 mmol.L-1 CaCl2 treatment reduced ABA content in the leaves of Sour jujube seedlings,increased GA3,IAA,ZT content and IAA/ABA,GA3/ABA,ZT/ABA ratio;5,10,20 mmol.L-1 CaCl2 treatment reduced ABA content in the roots of Sour jujube seedlings,increased GA3,IAA content and IAA/ABA,GA3/ABA ratio in the roots of 5,10,20mmol.L-1 CaCl2 treatment,and ZT content in the roots of Sour jujube treated for 12 hours.The ratio of ZT/ABA in roots treated with 20 mmol.L-1 CaCl2 was lower than that of control after48 hours.2.By transcriptome sequencing of Sour jujube seedlings:Compared with the control group,under exogenous CaCl2 treatment,7365 and 1247 differentially expressed genes were up-regulated and down-regulated in the leaves of wild jujube seedlings,5623 and 762 in the roots,respectively.GO database comparison showed that 43 GO conditions were enriched in the leaves of jujube seedlings,42 GO conditions were enriched in the roots,and the catalytic activity of leaves and roots was enriched to the most differential genes.KEGG database comparison showed that the differentially expressed genes were mainly involved in plant pathogen interaction,MAPK signaling pathway-plant,plant hormone signal transduction,phenylpropanol biosynthesis,pentose and glucuronic acid conversion,etc.
