In nature,the plant is always in the face of all sorts of pathogen attack,in the long-term cooperative coevolution,plants produce two levels of the immune system to resist pathogens invasion,the first is known as the innate immune system,immune system by pathogenic microorganisms associated molecular patterns PAMPs excitation,and after identification of plant cell surface receptor;The second kind of immune system is called effector-triggered immunity(ETI).Elicitors can simulate the interaction between pathogens and plants and stimulate the innate immunity of plants.Chitosan is an activator of chitin partial deacetylation,which can induce plants to resist fungal,bacterial and viral infection.In Arabidopsis,Chitosan regulate stomatal movement through the salicylic acid pathway,but the mechanism by which Chitosan transmit signals and how they affect plant defense is still unclear.In the previous study,transcriptome analysis and bioinformatics analysis of N.benthamiana treated with chitosan showed that some genes responded to chitosan expression,and then virus-induced gene silencing(VIGS)was used.Functional analysis of the candidate genes.it was found that NbSCF1 may be involved in plant defense response as a tobacco thioredoxin in tobacco.The localization analysis revealed that it was on the cell membrane.On this basis,NbSCF1 highly efficient silencing plants were obtained,It was found that NbSCF1 silenced plants promoted Chitosan-induced stomatal closure.ROS and NO staining revealed that NbSCF1 silencing increased Chitosan-induced ROS and NO accumulation,and NbSCF1 silenced plants enhanced chitosan-induced resistance to Sclerotinia sclerotiorum.Staining revealed that NbSCF1 silencing increased chitosan-induced H2O2 accumulation,and aniline blue staining showed that NbSCF1 silencing increased chitosan-induced sputum accumulation.In summary,NbSCF1 induces stomatal closure by regulating the effects of ROS and NO accumulation,and NbSCF1 regulates resistance to Sclerotinia by H2O2 and enamel.The results complement the chimeric Chitosan-induced plant immune signaling.
