This paper study on the effective parts of Chinese medicine compound,It is the core content to explore the material basis of pharmacodynamics of compound.Yupingfeng polysaccharide(YPF-P)is an effective part of Yu ping feng san.The previous study of our group has been confirmed YPF-P have Immuno suppressed mice and extensive immunomodulatory effects.However,the morphological and functional effects of YPF-P on intestinal mucosal immunity and the regulation of intestinal mucosal immune mechanism are rarely reported.The study on the mechanism of intestinal mucosal immune function has not been carried out.Therefore,the research in this paper has important theoretical significance and clinical value,the research results have a broad application prospect.First of all,this experiment use water extraction and alcohol precipitation method to extraction YPF-P and the compound of Astragalus polysaccharides?atractylodes macrocephalaon polysaccharides and Saposhnikovia divaricata polysaccharides,to explore Compound effect of YPF-P on intestinal morphology and immune function:80 SPF mice were randomLy divided into YPF-P group,polysaccharide group of Atractylodes macrocephala,astragalus polysaccharide group,wind polysaccharide group and blank control group.Different polysaccharides(200mg/Kg)and normal saline were given respectively.After continuous administration of 7d,they were sacrificed to death.The changes in the morphological structure of the small intestine were observed and the number of related immune cells was counted.The changes of IL-6,IL-2,TGF-beta 1 and secretory immunoglobulin A(SIgA)secreted by the small intestine were detected by ELISA,so as to explore the effect of YPF-P on the morphological structure of small intestine and the effect of intestinal immune function on the mice.Last,the effects of YPF-P on intestinal mucosal immunity in immuno-suppressed mice:80 mice were randomLy divided into blank control group,immuno-suppressive group and low,medium and high dose YPF-P group.Except for the blank control group,the other four groups were intraperitoneally injected with cyclophosphamide 80mg/Kg to replicated the immunosuppressive model.The low,middle and high dose groups were administered the YPF-P of 100,200 and 400mg/Kg respectively,and were given 7d after continuous administration.First,the blood routine,body mass andspleen index of mice were detected,and the model was successfully established.The changes of the morphological structure,immune function and the expression of various apoptotic factors were observed,and the regulation and mechanism of YPF-P on the intestinal mucosal immune response were investigated.Astragalus polysaccharides,Atractylodes macrocephala polysaccharides and Radix polysaccharides can be used in different ways increaseintestine of miceIntestinal villi length,V/C,The number of related immune cells can enhance the integrity of intestinal morphology and intestinal mucosal immune function in mice.The compound effect of Yupingsan,which is composed of the combined compound,is more significant.In immunosuppressive mice,YPF-P can relieve and improve intestinal mucosal immune damage induced by cyclophosphamide.,and can By adjustment Bcl-2/Bax?P53/Caspase3? TNF-? Expression regulation Apoptosis of intestinal epithelial cells protects intestinal mucosal immune function in immunosuppressed mice.
