| The sika deer(Cervus nippon hortulorum)is a kind of protected animal of the national grade I,which is a rare deer in China.The roe deer(Capreolus pygargus)is an important economic animal in China and one of the most extensive species of deer.In the northern mountains of Changbai Mountain,both of them are the main prey of the Amur Tigers and the Amur Leopards.The genetic adaptability of sika deer and roe deer in the same domain distribution has always been a hot issue of current biodiversity conservation concerns.We collected 104 samples of suspected wild sika deer and wild roe deer in the Changbai Mountain area of the Wangqing Forestry Bureau in Jilin Province and the Suiyang Forestry Bureau in Heilongjiang Province from November 2017 to March 2018.The DNA was extrscted by Qlamp DNAstool Mini Kit in stool DNA Kit.This study used the mitochondrial cytochrome b gene to identify species of suspected wild sika deer and wild roe deer collected in northeastern China,to identify whether they belong to sika deer or roe deer,and to use microsatellite markers for wild sika deer and wild roe deer for identified species.The samples were individually identified,and the differences in genetic diversity between wild sika deer and wild roe deer were analyzed by microsatellite.The results were as follows:1、The Blast alignment of the mitochondrial cytochrome b gene sequence was used to determine the species source of the fecal samples.It was determined that only 24 samples of the fecal samples were sika deer stool samples,and the rest were roe deer stool samples.2、The 12 pairs and 7 pairs of microsatellite primers were used to identify individuals between sika deer and roe deer by genotyping.It was determined that 24 sika deer stool samples were from 21 individuals and 80 roe deer stool samples were from 69 individuals.3、The microsatellite analysis of the genetic diversity of sika deer and Siberian roe deer was found that:The genetic diversity of sika deer and Siberian roe deer was higher,the genetic diversity of roe deer(He=0.8044)was higher than that of sika deer(He=0.7806).And the genetic diversity of the Northeast Sika deer has recovered from recent years,and the genetic diversity is higher than that of 2013(He=0.721),2017(He=0.730). |
