Transcriptome Analysis Of Different Compatibility Of 'Original Love' Ś 'Tiny Skyline'/'White Fox' In Lilium And Hybrid Identification

Lily is a perennial herbaceous bulb flower of Liliaceae.It is one of the most important bulbous flowers and cut flowers in the world.In order to explore the hybridization compatibility of odd-allotetraploid lily diploid Asian lily and Longiflorum lily,the odd-allotetraploid lily ?Original Love?(LAAA)was used as the female parent,diploid Asian lily cultivar ?Tiny Skyline?(AA)and longiflorum lily cultivar ?White Fox?(LL)used as male parent.We used genomic in situ hybridization to analyze the chromosome composition of the female and its progenies.And the hybridization compatibility was analyzed at the molecular level by transcriptome technology.The results were as follows:1.The results showed that the fruits of LAAA Ś AA were well developed,and 65 progenies were obtained through embryo rescue,while those of LAAA Ś LL aborted.Genomic in situ hybridization results showed that the ?Original Love? was near odd-allotetraploid(2n=4x=48).It includes 7 Longiflorum chromosomes 37 Asiatic chromosomes and 4 recombinant chromosomes.The chromosome numbers of the five progenies was about 48.Two of them were allotetraploid,the other three were aneuploid,which had different recombinant chromosomes.The results showed that ?Original Love? had a better fertility when using as female parent and it could play an important role in lily introgression breeding.2.The differentially of expressed Unigenes in the ovaries and leaves of LAAA Ś AA and LAAA Ś LL were analyzed by transcriptome sequencing ten days after pollination.And the unpollinated ovaries were used as a control.The results showed that:(1)There were 659 unigenes(446 up-regulated,213 down-regulated)in the ovaries of LAAAŚAA and 8,676 unigenes(6,615 up-regulated and 2,061 down-regulated)in the ovaries of LAAAŚ LL.There were 4240 unigenes(2737 up-regulated,1503 down-regulated)in leaves of LAAAŚAA,and 1548 unigenes(595 up-regulated,953 down-regulated)in leaves of LAAAŚLL.It can be seen that the number of unigenes in the ovaries of LAAAŚAA was less than that of LAAAŚLL,but the number of unigenes in the leaves of LAAAŚAA was more than that of LAAAŚLL.The two hybrid combinations of ovaries and leaves of unigenes showed diferent trends.Eighteen differentially expressed genes(9 came from ovaries and 9 camefrom leaves)were randomly selected for expression analysis by qRT-PCR.The results were consistent with the results of transcriptome sequencing.(2)In the ovaries and leaves of LAAAŚAA,the unigenes related to plant hormones such as auxin,cytokinin,gibberellin,antioxidants,etc.were up-regulated.But these unigenes are less expressed in the ovaries and leaves of LAAAŚLL.However,in the ovaries and leaves of LAAAŚLL,the expression of unigenes related to abscisic acid,ethylene,jasmonic acid and salicylic acid were up-regulated,while they were not expressed in the ovaries and leaves of LAAAŚAA.In this study,many unigenes related to auxin,such as SAUR in ovaries,ARF and TIR1 in leaves,were up-regulated in ovaries and leaves of LAAAŚAA.But the related unigenes changed little in the ovaries and leaves of LAAAŚLL.Antioxidant related unigenes,such as synthesizing SOD,PPO,POD were down-regulated in leaves of LAAAŚLL,but the glutathione metabolism and carotenoid biosynthesis unigenes were up-regulated in leaves of LAAAŚAA.This indicated that auxin and antioxidants played an important role in LAAAŚAA.
