Lily,a world-famous ornamental flower,is a perennial bulbous plant of the genus Lilium of the family Liliaceae.At present,the breeding of lilies in China is progressing slowly.There is a lack of commercial lily cultivar developed independently.Lily bulbs had always relied on imports and the breeding of edible lily was rarely reported.Therefore,it is of great significance to breed new cultivars with both good ornamental and edible value.In this study,L.brownii var.viridulum and L.davidii var.unicolor were selected as experimental materials.Chromosome Preparation and FISH were used to observe the chromosomal behavior of pollen mother cells during meiosis.L.brownii var.viridulum and L.davidii var.unicolor were carried out by cutting style pollination and embryo rescue.In situ hybridization(GISH and FISH)was used to analyze parents and hybrid progenies.The scale propagation,stomatal density,chlorophyll content,soluble protein content,total soluble sugar content,and starch content of the F1 progenies were measured.Results are as follows:(1)The meiosis of L.brownii var.viridulum and L.davidii var.unicolor was basically normal,with little abnormalities such as chromosome malsegregation and tetrad.The pollen germination rates of L.brownii var.viridulum and L.davidii var.unicolor were31.26%and 45.56%,respectively.(2)The compatibility of the obverse and inverse crosses of L.brownii var.viridulum and L.davidii var.unicolor were significantly different.When L.brownii var.viridulum as the female parent and L.davidii var.unicolor as the male parent,98 fruits were obtained form 140 flowers pollinated with cut style with a maturing rate of 70.0%,and 166 seedings were obtained through embryo rescue and the emergence rate was 5.6‰.However,there were no developed fruits with conventional pollination.No fruits were obtained through normal or cut style pollination,when L.davidii var.unicolor as the female parent and L.brownii var.viridulum as the male parent.(3)The results of in situ hybridization showed that both the parents and the hybrid progenies were diploid(2n=2x=24),and they had a genome of L.brownii var.viridulum and a genome from L.davidii var.unicolor.(4)Analysis of physiological traits of parents and F1 progenies showed that:the proliferative coefficient of some F1 progenies were higher than that of the parents,and most of them were between the parents;the stomatal density of some F1 progenies were not significantly different from that of parents,and others were significant difference between parents;The chlorophyll content of some F1 progenies were higher than that of the parents,and most of them were not significantly different from the male parent L.davidii var.unicolor;L.brownii var.viridulum had the highest soluble protein content,and the differences in the contents of F1 progenies were large;the total soluble sugar content of the hybrid progenies were higher than parents;the F1 progenies and parents generally had low starch content.The above research results showed that when the female parent was L.brownii var.viridulum and the male parents was L.davidii var.unicolor,hybrid progenies could be obtained by cut style pollination and embryo rescue.Physiological indicators of parental and hybrid progenies seedlings analysis found that there was a large difference between the hybrid progenies and the parents,which provided more possibilities for the selection of new lily cultivar and could initially screen high-quality hybrid progenies.This study had laid a foundation for breeding new edible lily cultivar with edible and ornamental. |
