Rationales: Radix Astragali is the dry root of the leguminous Astragalus membranaceus Bge.var.mongholicus.Hsiao and Astragalus membranaceus.With the improvement of the awareness of universal health care,the market demand for jaundice has increased rapidly,and the wild sassafras resources have been unable to meet the market demand.The cultivated Astragali Radix has been developed on a large scale.The key to ensuring the quality of medicinal materials and the safety and efficacy of traditional Chinese medicine is to ensure the stability and uniformity of the quality of medicinal materials.The chemical components in medicinal materials are the material basis for exerting efficacy,and are susceptible to various factors such as species,origin,ecological environment and growth mode.Variation occurs due to influence [1].However,the medicinal materials are classified and used only when the chemical composition of the medicinal material is changed to cause a change in the medicinal effect.Among the many influencing factors,the compositional changes caused by the planting method are larger than the species and the producing area [2].At present,the commodities circulating in the market are mainly Mongolian Astragalus,and the main producing areas of Mongolian Astragalus are Shanxi,Inner Mongolia,Shaanxi,Gansu and other places.Gansu is mainly based on transplanting Mongolian scutellaria,while Shanxi and Inner Mongolia are mainly wild-type.The wild-type Mongolian scutellaria can generally grow for more than 5-6 years,while the transplanted Mongolian scutellaria has a growth period of 2 years.Studies have shown that there are differences in appearance traits,chemical composition,and efficacy.According to the central law,genetic material DNA is determined by determining the appearance traits and chemical components.Therefore,we use ISSR molecular marker technology to study the genetic diversity of different growth patterns and ages of Mongolian scutellaria,in an attempt to find DNA related to growth patterns and years.The band establishes the basis for the subsequent study of the correlation of secondary metabolites of Astragalus.Objective: In this study,in the similar climate near the Hengshan Mountains in Shanxi Province,1-6 years of wild Mongolian scutellaria and 1-2 years of transplanted Mongolian scutellaria were selected.The effects of different growth patterns on the genetic diversity of Astragalus membranaceus were studied under the same growth mode,different growth years and the same growth years.And to find the DNA bands related to the growth mode,the number of years,for the subsequent study and the basis of the secondary metabolites of Astragalus membranaceus.Methods: ISSR labeling technique was used to amplify the Astragalus membranaceus.The genetic map of Astragalus membranaceus was constructed by POPGENE 1.32 and NTsys-pc software.Correlation analysis was carried out to find out the relevant bands with different growth environments and different ages.Result: Mongolian Astragalus membranaceus was rich in genetic diversity by analyzing the genetic diversity index P<58,PPB=98.31%,Ne=1.7178,He=0.3954,I=0.5738,Ht=0.3954.Using the correlation analysis,four ISSR bands related to growth years were found,855-3*,827-4*,and two bands related to growth patterns,840-2*,866-3*.Conclusion: Through the genetic diversity index of 1-6 years old Mongolian scutellaria,P=58,PPB=98.31%,Ne=1.7178,He=0.3954,I=0.5738,Ht=0.3954,and the genetic diversity of 1-2 year old transplanted Mongolian scutellaria The index has Na=1.9180,Ne=1.6039,He=0.3471,I=0.5126,P=56,PPB=91.8%,Ht=0.3471 analysis,indicating that Mongolian scutellariae is rich in genetic diversity,and the genetic diversity of wild Mongolian scutellaria The sex level is higher than that of transplanted Mongolian scutellaria.The 1-6-year-old Mongolian Astragalus membranaceus was clustered into three groups when the genetic similarity coefficient was 0.612.I gathered a variety of years of jaundice;II gathered some of the 1,3,and 4-year-old jaundice;III gathered 4,6-year-old part of jaundice.1-2 years old transplanted Mongolian scutellariae clustered into 2 classes when the genetic similarity coefficient was 0.601,1 year in class I,and half in 2 years;in class II,it was roughly 1 year old,and 2 years old jaundice was first grouped together.Then gather in a big class.Using SPSS correlation analysis,two ISSR bands related to growth years were found,855-3*,827-4*,and two bands related to growth patterns,840-2*,866-3*.
