The phenomenon of rice albinism is widespread in the plant kingdom and its mechanism is very complex.Studies have shown that rice albinism is affected by environmental factors such as light,temperature and mineral elements,and is also related to genetic mutation,karyoplasmic interaction and the expression of genes related to chloroplast and chlorophyll synthesis.In recent years,the mechanism of rice albinism has been studied more and more deeply.The main sources of rice albino mutants were natural mutation,Ethyl methanesulfonate(EMS)mutagenesis and 60Co irradiation.However,there are few reports about the albino vaccine caused by agrobacterium infection.In this study,the albino seedlings and normal seedlings of guihefeng,an indica rice variety,were obtained by Agrobacterium-mediated in Planta of rice.Then the differentially expressed genes involved in chloroplast synthesis,light and pigment synthesis,photosynthesis and other metabolic pathways were explored by transcriptome sequencing technology.The preliminary results are as follows:1.In situ transformation of rice seeds infected by Agrobacterium tumefaciens,the albino seedling rate was more than 69%.2.The growth of albino seedlings and normal seedlings of rice were compared and analyzed.The results showed that albino seedlings of rice grew slowly and their roots were underdeveloped.3.The ultrastructure of albino leaves and normal leaves of rice showed that the cause of albino leaves was related to chloroplast deficiency.4.The activities of antioxidant enzymes and the content of MDA in rice leaves and roots were determined respectively.The results showed that the activity of antioxidant enzymes increased and the content of MDA increased significantly,which indicated that the active oxygen species increased and the lipid structure of cell membrane was destroyed during in situ transformation of rice.5.Transcriptional histology of albino and normal leaves was analyzed by Illumina sequencing platform.A total of 42031 genes were detected and 1024 differentially expressed genes were screened out.Two differentially expressed genes related to chloroplast synthesis and 11 differentially expressed genes related to photosynthetic carbon fixation,flavonoid biosynthesis,chlorophyll metabolism and carotenoid biosynthesis were identified by further bioinformatics analysis.At the same time,some differentially expressed genes encoding AP2/ERF,NAC and MYB transcription factors were also found.6.Using actin6 as internal reference gene,real-time fluorescence quantitative verification was performed on 15 randomly selected differentially expressed genes that might be related to chlorophyll synthesis.The results showed that transcriptome sequencing was reliable.
