The widespread spread of citrus Huanglongbing has caused serious economic losses,and its pathogens(CLas)are currently unable to achieve laboratory culture,leading to rare breakthroughs in research progress.Citrus psyllid is the only natural insect vector of Huanglongbing.This study identified a population of citrus psyllids and established a group of granulated sugar granules as a sterile sugar orange seedling in the greenhouse.Orange research model.From the model,an absolute quantitative method was used to screen three samples of granulated orange seedlings carrying high,medium and low concentrations of Huanglongbing and a set of healthy control granulated orange seedling samples.Four groups of samples were quickly extracted by real-time direct analytical mass spectrometry(DART-MS)and analyzed by omics.The specific research progress and results in this study are summarized as follows:A population of citrus hibiscus with a prevalence rate of 56.67% was identified.The research model of the succulent orange was established by bagging the inoculated sugar orange seedlings.The preliminary time rule of the sucrose orange seedlings from the citrus hibiscus was found.The minimum incubation period was 22 days,the median latency was30 days,and the sustained retention period was 46 days.By principal component cluster analysis of the sugared orange and sterile sugar orange metabolomes,it was found that there was a significant difference between the bacteria and the sterile group,and the samples with low concentration of Huanglong pathogen were compared with the high and medium concentrations of the healthy samples.The difference of the samples was more significant.The target difference compounds of 252 bacteria samples and sterile samples were compared in METLIN data by statistical screening mass spectrometry.The differential compounds were matched by metabolic pathways,and 10 different compounds were identified.In the metabolic pathways;the first 10 identified compounds are: Phytosphingosine,Spirilloxanthin,4-methyl-Methlfecosterol,Table Episterol,Estrone,D-Cathinone,L-Pipecolate,N-Succinyl-L-glutamate,L-valine(L-Proline),Tetrahydrobiopterin;the six types of metabolic pathways that are matched are:ester metabolism,terpenoid/polyketide metabolism,steroid/steroid hormone synthesis Degradation and metabolism,amino acid metabolism and biosynthesis of other secondary metabolites.The main research results and conclusions in this paper are of great significance to production practice and theory,and help to analyze the pathogenic mechanism of huanglongdisease,and also provide a feasible reference for the development of new diagnostic technology of huanglongdisease by marker compounds..
