To investigate the prevalence of canine mammary tumors,we collected canine samples from several animal hospitals from 2016 to 2018,and studied the clinical symptoms, disease types, ages and daily diets, etc. The results showed that the incidence of canine mammary tumors in poodle, dachshund and pomeranian was 39.79%,9.18%and 9.18%,respectively.The ages ranged from 3 to 16 years old, including 26 cases under 9 years old, accounting for 26.5%,and 64 cases between 9-13 years old, accounting for about 65.3%,8 cases over 13 years old, accounting for about 8.1%,the disease mostly occurred in female dogs aged 9-13.The pure dog food plus meat is the major diets for canine tumors, and this dietary structure is the highest incidence of canine mammary tumors with a percentage of 58.16%.In veterinary clinical treatment, surgical removal is a common method for canine mammary tumors.Histopathological identification is the gold standard for benign and malignant tumor identification.50 cases of canine mammary tumors were removed by surgical method, and were analyzed by H.E.staining.The results showed that surgical is the most common treatment method. In 50 cases,30 cases were benign tumors, including 15 simple adenomas,8 mixed tumors,5 composite adenomas,1 fibroid, and 1 fibrous adenoma.20 cases were malignant tumors, including 18 simple cancers and 2 composite cancers.Among the 30 benign tumors,15 were simple adenomas, including 9 cases of ductal tumors, accounting for 60%,1 case of ductal papilloma, accounting for 6.67%,and other types of simple types, accounting for 33.33%.Among 20 malignant tumors,18 were simple cancers, including 2 cases of ductal papillary carcinoma, accounting for 11.11%,6 cases of ductal carcinoma, accounting for 33.33%,7 cases of parenchymal cancer, accounting for 38.89%and other types of simple cancer, accounting for 16.67%.To investigate the relationship between expression of Oct-4 and development of canine mammary tumors,50 cases of canine mammary tumors were examined by immunohistochemistry. The results showed that the positive expression of Oct-4 were mainly around the cell nucleus. In 50 tumor cases,28 cases had strongly positive expression of Oct-4,17 cases had moderate positive expression, the positive expression rate was 90.0%(45/50).The expression of Oct-4 was significantly different between tumors and normal tissues(p<0.05).Among the 50 immuno-histochemical tumors,14 of 30 benign tumors had strong positive expression of Oct-4,the rate was 46.67%,11 cases had positive expression, the rate was 36.67%.14 of 20 malignant tumors had strong positive expression, the rate was 70%,and 6 cases had positive expression, and the rate was 30%.The results showed that the occurrence of canine mammary tumors was closely related to the breeds, ages and diets. Most of the tumors obtained were benign, and the simple adenomas were the major.Furthermore,Oct-4 was strongly positive in tumor cells.
