Leymus chinensis is a perennial quality forage that is nutritious.Leymus chinensis has good palatability,high yield and high feed value,but its own “three lows” problem limits the yield of Leymus chinensis seeds.How to break seed dormancy and improve the germination rate o f Leymus chinensis seeds is the key to the production of Leymus chinensis.In this experiment,we compared the effects of different chemical agents and hormones?GA 3?on the germination rate of Leymus chinensis seeds,determined the best germination treatment plan,and investigated the molecules of GA3 promoting the seed germination of Leymus chinensis by high-throughput sequencing.mechanism.The specific results are as follows:?1?Through the determination of the final germination rate,germination index and germination potential of Leymus chinensis and Leymus chinensis seeds,the germination effect of various treatments was different,but the germination rate of Leymus chinensis seeds was generally lower than that of Leymus chinensis seeds..The germination rate of Na OH and H2SO4 seed soaking treatment group was lower than that of the control group.The germination rate of lye + GA3 composite seed soaking treatment was 1.25 mol/L Na OH+100 ug/g GA 3<CK<1.25 mol/L.Na OH+300 ug/g GA3<1.25 mol/L Na OH+200 ug/g GA3;in GA3 treatment,the germination rate was 100 ug/g GA3<300 ug/g GA3<200 ug/g GA3.Overall,the highest germination rate was treated with GA3 at a concentration of 200 ug/g,and the germination rate was 83.29%.?2?In this experiment,three different treatments were applied to the leech and the sorghum seed of the Leymus chinensis seeds,ie,immersion in CK and distilled water soaking treatment for 24 h,germination for 72 h and 200 ?g/g GA3 soaking for 24 h and then germination.At 72 h,a total of six samples were sequenced by transcriptome.A total of 37208 single gene clusters?Unigenes?were obtained by screening and assembling the measured sequence fragments.The length was mostly distributed between 200 and 300 bp,and the length of N50 was 1541 bp.?3?Under the condition of significant level p?0.05 and fold change?2,compared with the treatment of distilled water soaking in GA3 soaking seeds of Leymus chinensis seeds,4309 significant differential genes were obtained,of which 2257 genes were u p-regulated and the gene was down-regulated.2052.Differential gene enrichment pathways are associated with starch and sucrose metabolic pathways,galactose metabolic pathways,phenylpropanoid biosynthetic pathways,alpha-linolenic acid metabolic pathways,abc transporter pathways,and photosynthesis antenna protein pathways.Among them,the most abundant pathways are the starch and sucrose metabolic pathways.?4?Under the same conditions,compared with the treatment of distilled water soaking,the GA3 soaking treatment of Leymus chinensis seeds resulted in a total of 11,919 differentially expressed genes,of which 8067 were up-regulated and 3852 were down-regulated.The pathways for differential gene enrichment are mainly related to ribosome pathway,phen ylpropanoid biosynthesis pathway,phagocytic pathway,cysteine and methionine metabolic pathway,oxidative phosphorylation pathway,cyanogenic amino acid metabolic pathway and phosphatidylinositol signaling system pathway.And so on.?5?In order to verify the accuracy of transcriptome data,10 genes were randomly selected and verified by real-time fluorescent quantitative PCR.The results were basically consistent with the up-regulation trend of transcriptome data.
