Leymus chinensis is one of the excellent grasses,which plays an important role in the restoration of degraded grassland,the construction of artificial grassland and the production of high quality forage grass.The chemical mutagenesis of ethyl methyl sulfonate(EMS)is one of the most widely used mutagenesis techniques.With low cost,high mutation frequency and abundant variation,EMS has been widely used in main crop breeding,such as wheat and rice.At present,there are few reports about the application of EMS mutagenesis on Leymus chinensis.The research not only can obtain abundant mutant materials for the subsequent research on gene function analysis and new variety breeding,but also be important to enrich the breeding technologies and methods.In this study,the seed of "Jisheng 4" varieties of Leymus chinensis was used as the materials,the effects of different EMS concentrations and treatment time on seed germination and seedling growth were studied to find the best treatment method for Leymus chinensis.Based on the phenotypic traits investigation and analysis,the mutants were screened.The variation patterns and mutation frequency were analyzed to reveal the characteristics of EMS mutagenesis of Leymus chinensis,and to support the subsequent research of EMS mutation breeding.The main results are as follows:1.EMS mutagenesis had a significant inhibitory effect on the seed germination of Leymus chinensis.The germination rate,relative germination rate,germination potential,relative germination potential,germination index and relative germination index of Leymus chinensis seeds decreased significantly with the increase of EMS mutagenesis concentration and mutagenesis time.Different concentrations and treatment time of EMS mutagenesis had significant delay effect on the seeds germination process of Leymus chinensis,and the initial germination time was delayed 2d-10d;EMS mutagenesis had a significant effect on the seedling growth of Leymus chinensis.The root length and seedling length decreased significantly after EMS mutagenesis.2.There was significant interaction on the mutation effect between mutagenesis concentrations and treatment time.The half lethal treatment of EMS mutagenesis on Leymus chinensis seeds was 1.5% EMS for 16 h.3.The M1 generation mutagenesis population was constructed with 643 individual plants.Based on the phenotypic analysis,120 plants with obvious phenotypic variation were obtained and the mutation frequency was 18.66%.Among them,the frequency of leaf color mutation was 15.09% and the frequency of leaf shape mutation was 3.27%.In addition,14 dwarf mutants,14 growtn-inhibited mutants,1 mutant with increased tiller number and 1 mutant with increased leaf were obtained.In this research,the effect of EMS mutagenesis Leymus chinensis was investigated and the optimized method was determined.The results will be helpful to construct the mutation bank which will be helpful for the discovery of novel functional gene and development of new cultivars.
