| The fine germplasm is the guarantee of the controllability and stability of the Traditional Chinese Medicine quality.However,it is significantly weak in Chinese Medicine breeding at present,which imperatively need us to do some creativity research from materials and methods.The hybrid varieties can provide fine quality,high yield,good resistive and stable germplasms for the medicinal plant production.And which attributes also can be fixed with asexual that has gained tremendously success in some Chinese medicine.But there is still no such a variety applied in the production in Bupleuri until to now.In this dissertation,we take the important Chinese Medicine(Bupleurum)as the subject to explore the feasibility of using heterosis in Buplrurum breeding.Which is to lay the foundation for the new varieties of Bupleurum.The results are as follows:1.Chromosome of F1 hybrids and its parentsFlow cytometry was used to identified the ploidy of F1 hybrids.The result showed that the ploidy of F1 hybrids and both parents were all diploid.Which manifested that F1 hybrids of Bupleurum attained in this present study has generation ability.2.Heterosis of F1 hybrids in yieldIn harvest period,root scanner and ruler were used to measure the root yield and other root indexes of F1 hybrids and its parents.The parameters all exhibited heterosis in the present study,but with different heterosis extent.The range of MPH for total six indexes was in 6.22 % ~ 82.01 %,and UPH in-7.99 % ~ 45.33 %.The maximum value of MPH was root length,which also showed UPH.And the rate of root dry to fresh weight was displayed negative UPH solely.The above results preliminarily confirmed that using heterosis was can significantly improve the yield of Bupleurum roots,showing fine application prospects.3.heterosis of F1 hybrids in Quality of three mainly componentsIn harvest period,HPLC was used to determine the contents of SSa and SSd,UV was applied in SStotal,total flavonoid,Soluble polysaccharide,reduce polysaccharide and total polysaccharide.There was exist tremendous differences among three mainly components in F1 hybrids.The range of MPH in seven indexes was from-47.62 % to 24.57 %.But MPH was just displayed in total flavonoid.The results illustrated that using heterosis in Bupleurum was an efficiency method to enhance the content of total flavonoid.4.Heterosis of F1 hybrids in growth adaptability(1).Morphology of F1 hybridsIn harvest period,leaf area analyzer and ruler were used to gained the statistics of F1 hybrids and its parents in morphology.Apart from LW,all parameters were exhibited heterosis with huge divergence.The range of MPH was from 5.18 % to 30.65 % among seven indexes,UPH from-22.96 % to 9.74 %.Meanwhile,the maximum of MPH was showed in PH accompanied with UPH,and the minimum in LA.What’s more,the emerging time of first node and first flowering(ETFN,ETFF)were all in advance compared with its parents.Above results preliminarily manifested that F1 hybrids showed fine growth adaptability in Sichuang.(2).Physiology in Photosynthesis and fluorescence of F1 hybridsIn vegetable and harvest period,respectively at April and December,portable photosynthesis system LI-6400,TYS-A chlorophyll meter,HK/ZYTYS-3N nitrogen metro and m-pea fluorescent metro were used to measure the indexes of photosynthesis and fluorescent physiology.The result showed that Pn,WUE,total Chl and PI abs were exhibited heterosis.The range of MPH was from 8.83 % to 31.13 % among four indexes,UPH from-22.96 % to 9.74 %.Meanwhile,the maximum of MPH was showed in PI abs,WUE accompanied with UPH,and the minimum in Pn.At harvest period,all indexes,apart from total Chl,displayed heterosis.The range of MPH was from 1.92 % to 17.60 %,UPH from-13.99 % to 2.38 %.The maximum of MPH was exhibited in WUE accompanied with UPH,and the minimum in PI abs accompanied with UPH.Besides,PI abs of MPH in vegetable period,was higher than harvest period to F1 hybrids(about 93.38 %),the photosystem efficiency and WUE of MPH were higher than vegetable period(about 41.90 %,42.53 % respectively).All above demonstrated that use the heterosis can significantly improve the growth adaptability of seedling and plants in Sichuang province.(3).Anti-oxidative ability of enzyme and extent of lipid in F1 hybridsIn harvest,UV was used to measure the contents of SOD,CAT and MDA.The result exhibited that SOD and CAT were showed positively heterosis,but MDA controversy.The result above were consistent with the results of morphology and photosynthesis and fluorescent.All above showed a basically results that F1 hybrids embrace fine adaptability in Sichuang province.According all the above results,in the present study was attained F1 hybrids crossed by Chuanbeichai NO.1 and Bupleurum margrintum.The analyses result of heterosis exhibited that the most of indexes for F1 hybrids were existed UPH,for instance,root yield as the economical index,Flavonoid contents as the quality index,PI abs as the growth adaptability index,which showed great application progress.According our study,the available of hybrids in Bupleurum was primarily proved,which made a foundation for F1 hybrids as a Chinese medicinal plant in the future.This research,what’s more,could have a hugely implication on sexual breeding,and which also was further abundant to the content of genetics in Chinese medicinal breeding. |
