Hainan Black Goats are valuable tropical germplasm resources,but their reproductive efficiency is low.In this study,the blood physical and chemical indicators and reproductive hormones of Hainan Black Goats were determined by measuring the reproductive cycle(anestrus,estrus,gestation,lactation),and the differences in physiological characteristics of the ewe in different reproductive stages were analyzed.The analysis provides theoretical basis;the iTRAQ technique is used to identify and analyze the serum proteome information of Hainan Black Goats in the breeding cycle,and to screen differentially expressed proteins related to reproductive traits,which provides scientific research on the management and expansion of Hainan Black Goats.The main results of the theoretical guidance are as follows:1.The reproductive cycle Hainan black goat blood physical and chemical indicators measured:white blood cells,hemoglobin,mean hemoglobin content and average hemoglobin concentration and other changes in blood physiological indicators,total bilirubin,alanine aminotransferase,triglycerides,cholesterol and other serum biochemistry The indicators changed significantly;it suggested that in the daily feeding management,attention should be paid to the anti-inflammation of the ewe,and the nutritional balance of the diet.The results of the blood reproductive hormone test showed that the FSH and LH secretion trends were consistent,and the secretion level in the estrus increased and entered the pregnancy.The lactation period continued to decrease;the secretion of E2 decreased after gestation,and the secretion gradually recovered after lactation;the secretion of PRL was not significant at all times;the secretion level of P4 during pregnancy was significantly higher than that of non-pregnant period;The law of the secretion of circulating reproductive hormones.2.iTRAQ identification and bioinformatics analysis showed that a total of 5745 peptides and 1213 proteins were identified,and differentially expressed proteins were screened by fold change>1.2 and Q value<0.05.The estrus/anestrus up-regulated 75 proteins,down-regulated 66 proteins;101 and 69 up-regulated and down-regulated proteins in gestation/estrus,respectively;58 up-regulated during lactation/gestation,down-regulated 101 proteins;further analysis of differentially expressed protein results,a total of five differentially expressed proteins such as APOC-? X2?C4BPa X4?FIX?LECT2?HABP2 X2 were screened out.The basic protein information between the various stages of Hainan Black Goats breeding cycle was preliminarily revealed,which provided a theoretical basis for the construction of Hainan Black Goats efficient breeding system.3.Pathway enrichment analysis of differentially expressed proteins showed that the pathways of libido/estrus,gestation/gestation,and the number of differentially expressed proteins in lactation/gestation were all complement and coagulation cascades.The maximal pathways of RF are the Fanconi anemia pathway(anestrus/estrus),adipocytokine signal transduction(anestrus/gestation),p53 signaling pathway(lactation/gestation);Pre-IGFBP2?ANTXR2?Ran?SFRP3??4GalT1?Pre-SAA21 and vWF seven differentially expressed proteins related to reproductive function,indicating that Hainan black goat ewes at different reproductive stages are different in many aspects of cell proliferation,adhesion and migration,nutritional metabolism,immune and inflammatory.
