The some kinds of epidemic of goats has been perplexing the development of Hainan black goat industry.In this study,a serological investigation of the existing Hainan black goat epidemics was conducted.The epidemiological types investigated were paratuberculosis,ORF,brucellosis,Clostridium welchii,pleuropneumonia and pasteurellosis.The investigation method is mainly based on the immunological reaction that is the antigen and the antibody specific reaction,which means ELISA.The results showed that there are paratuberculosis,ORF,Clostridium welchii,pleuropneumonia and pasteurellosis in Hainan black goat population,but no brucellosis.Through a field survey of Hainan black sheep,we found that pneumonia related diseases persisted in flocks.In order to further discover the pathogenic bacteria of Hainan black goat pneumonia-related epidemics,this study based on the clinical symptoms of the black goat,successfully isolated and identified Pasteurella multocida type D and Burkholderia pseudomallei Hainan strain by culture,biochemical identification and molecular biology methods.Then in order to improve the prevention and control measures of Hainan black goat against Pasteurella multocida type D,this study carried on the drug susceptibility test to this pathogen,and found that is sensitive to trimethoprim-sulfamethoxazole,levofloxacin,tetracycline,Taicangmycin and amoxicillin,moderate sensitivity to ceftazidime and erythromycin and resistance to florfenicol.Finally,based on the high pathogenicity of Burkholderia pseudomallei and its resistance to most antibiotics,this study extracted the whole genome of the bacterium and successfully obtained the whole genome of Burkholderia pseudomallei Hainan strain sequencing using the Illumina Hiseq 4000 platform combined with the Pacbio RSII platform,and And perform genome annotation,resistance gene analysis,and virulence gene analysis.This can provide genomic information support to the establishment of a standard strain of Burkholderia pseudomallei in China,constructing an animal model of infection and providing an effective vaccine as early as possible.
