In the application of biological control,using natural enemy to control pest is an important method.Arma chinensis is a predatory natural enemy with great potential exploitation and can control various agricultural and forestry pests of lepidoptera,coleoptera,hymenoptera and hemiptera.However,in practical applications,the massive propagation and establishment of Arma chinensis has been an important factor affecting its function.In order to further understand the nutrition,In this research,we sequence genomic and salivary glands transcriptome sequences of Arma chinensis,Then we made a series of molecular biology of its fatty acid metabolism based on omics data,This study provides scientific and theoretical basis for the research on the nutrition mechanism of Arma chinensis.Genomic sequencing of Arma chinensis was conducted in this study,PCR-free SMRT bell library was constructed,and 128 G of original sequence was obtained by sequencing with PacBioSMRT Technology.After assembly and splicing,the 983 Mb genome sequence of Arma chinensis was obtained.Its Conting N50 was 2.39 Mb,and 17681 genes with annotation function were found by bioinformatics analysis.Based on genomic annotation information of 5 insect species including Arma chinensis salivary glands transcriptome and genomics,241 genes related to wing formation and movement and 67 genes related to flight were homologously compared,among which 66 genes related to myosin.Two genes expressing bursicon and four troponin C genes were expressed.In addition,there were 305 zinc finger protein genes,24 genes related to fat metabolism in digestion,and 1 gene expressing fatty acid hydroxylase.In this study,the influence of inbreeding on the growth and reproduction of Arma chinensis was measured and found that:Inbreeding breeding and cross breeding of Arma chinensis egg hatchability and incubation duration has significant differences,hatching rate(66.28%)of the hybrid breeding was22.62% higher than inbreeding reproductive system(43.66%),The incubation of hybrid breeding and inbreeding reproductive also has extremely significant difference,inbreeding breeding egg incubation period(7.70 d)was 1.67 d longer than hybrid breeding system(6.03 d),and on the stability,inbreeding breeding are significantly lower than the hybrid breeding.After six generations of inbreeding,Arma chinensis population shows male-biased phenomenon.However,there were significant differences in the average single spawning number between the two systems.The crossbreeding(26.60 grains)was4.29 times higher than the inbreeding(22.31 cycles).However,there was no significant difference in body weight,developmental history and survival rate of nymphs at different inbreeding ages.We sequenced the salivary glands of Arma chinensis and Halyomorpha halys by transcriptome sequencing.Orthologous genes through the transcriptome data analysis,screening 55 rapidly evolving gene,the expression of fatty acid hydroxylase(FAXDC2)unigene has important role for fatty acid oxidation of alpha.The gene was verified by qPCR,found in the salivary glands of the two is not significant difference,however significant difference expressed in midgut,The expression level of Arma chinensis in its midgut is significantly higher than it in Halyomorpha halys midgut.It indicates thatFAXDC2 may play an important role in the digestion of Arma chinensis,and its active site is mainly in the midgut.This effect needs further experimental verification.
