Objective:To provide an efficient genetic diagnostics method for Labrador Retrievers and Golden Retrievers,15 SNPs of 8 canine hip dysplasia(CHD)related genes in genomic DNA of Labrador Retrievers and Golden Retrievers were detected.Methods:1.30 normal and 25 typical CHD Dogs were selected by imaging diagnosis.2.The vein blood was sampled and the genomic DNA was isolated from white blood cells.3.The PCR/sequencing method was used to analyze the variants of 15 SNPs.4.Analysis and calculation of the percentage of parting,differences in normal dogs and hip dysplasia.Results:1.The variants of COL6A3(CFA25:51040259)significantly associated with CHD in Labrador Retrievers(P=0.013),the A allele frequency of CHD samples is significantly higher than normal sample(P=0.006).The variants of FN1(CFA37:25095511)significantly associated with CHD in Labrador Retrievers(P=0.003),the A allele frequency of CHD samples is significantly higher than normal sample(P=0.001).The variants of COL1A2(rs22356416)is significantly associated with CHD in Labrador Retrievers(P=0.032),the C allele frequency of CHD samples is significantly higher than normal sample(P=0.023).The variants of COL1A2(rs850558710)appear to associated with CHD in Golden Retrievers,but not significant(P>0.05),the A allele frequency of CHD Golden Retrievers samples is significantly higher than normal sample(P=0.039).2.COL6A3(CFA25:51031100),LAMA(CFA1:70997779),SCR(CFA24:28944481),LRR1(CFA8:29247021),DLX3(rs851998705,rs851998705),GDF5(rs8821566,rs851464214)and COL1A2(rs850833341,rs852724798,rs851504841,rs852954163)SNP loci were not significantly associated to CHD.Conclusion:The variants of COL6A3(CFA25:51040259),FN1(CFA37:25095511),COL1A2(rs22356416)is significantly associated with CHD in Labrador Retrievers(P<0.05),These SNPs could provide an efficient genetic diagnostics method for Guide Dogs.
