Objective:1. To establish the method for detecting SNP loci of coat color gene MC1R(c.C916T) and TYRP1(c.C991T) in canine.2. To investigate the relationship between coat color, the genotypes of coat color, sex of Labrador Retriever and Golden Retriever and the training success rate of guide dogs.Methods: In the study, the dogs and the information of the dogs were provided by Chinese Guide Dogs Training Centre of Dalian; the dogs that detected in the study were identified and assessed by the identification personnel according to the mature identification method. The polymorphism of the SNP loci of coat color gene MC1R(c.C916T) and TYRP1(c.C991T) of45 Labrador Retrievers and 16 Golden Retrievers were detected and analyzed. And the genotypes of coat color of each dog were typed, to discuss the correlation between coat color and genotypes of coat color of dogs and the training success rate of guide dogs.Results:1. The results show that, PCR/ sequencing method can detect the SNP loci c.C991 T of gene TYRP1 clearly and effectively, but the sequencing result of the SNP loci c.C916 T of the MC1 R gene appeared a special peak, which was finally confirmed as heterozygosity SNP loci by cloning / sequencing method.2. 16 Golden Retrievers(14 male 2 female)were detected, the coat color phenotypes were all golden, coat color genotypes were all ee BB genotypes without others; 45 Labrador Retrievers(23 female 22 male) were detected, the coat color phenotypes were yellow(23) and black(22), coat color genotypes were ee BB(4), ee Bb(19) yellow dogs, EEBb(3),Ee BB(14),Ee Bb(4)black dogs, no other coat color genotypes like EEBB and EEBb(black),eebb(yellow)were detected. Besides, because the chocolate labrador retriever are rare, and there is no chocolate labrador retriever here, so the corresponding genotypes of Eebb and EEbb were not detected either.3. Using correlation analysis of χ2 test, there was no significant correlation between coat color and coat color genotypes of Labrador Retriever and Golden Retriever and the training success rate of guide dogs(P > 0.05); in addition, there was no significant correlation between the sex of Labrador Retriever and Golden Retriever and the training success rate of guide dogs(P >0.05).Conclusion: There is no significant correlation between coat color, coat color genotypes and sex of Labrador Retriever and Golden Retriever and the training success rate of guide dogs,they can’t be the effective index of selecting guide dog candidates to prove the training success rate of guide dogs.
