Dermatophytes,a polymorphous filamentous fungi,infect protein in the skin and subcutaneous as well as a highly pathogenic bacteria both to skin infections in human beings and animal.chemically-synthesized medicine and hormone were widely used to cure dermatomycosis clinically,however,their induces the drawbacks like side effects and drug-resistant strains.The results according to pre-experimentes showed that Sophora flavescents Dictamnus dascarpus,Cortex Pseudolaricis,Stellera chamaejasme L and propolis,commonly used in traditional Chinese medicine usage,possess strong inhibition effect on Dermatophytes.therefore,five kinds of Chinese herbal medicines were decided as the object of study.Under the modern extraction methods,the active ingredient was obtained.The antifungal activity in vitro of five herbal extracts on Microsporum canis,Trichophyton mentagrophytes and Trichophyton rubrum was investigated on the the CLMI M38-A2.Evaluation efficacy of the compound Chinese medicine was conducted on animal models.The experiment on stimulation and security of self-made compound antifungal reagents.Results 1:The active ingredient of Sophora flavescents Dictamnus dascarpus,Cortex Pseudolaricis,was obtained through Condensation reflux extraction.Extraction rate and antifungal activity of the extractions from the five Chinese medicine were investigated by M38-A2 broth microdilution method.The result showed that for the three Chinese medicines,the ratio of ethanol extractives is 4.317%、7.803%and 9.162%,which was twice as high as the ratio of water extractives and the minimal fungicidal concentration(MFC)of ethanol extractives is lower than the water extractives.The ethanol extractives of the five medicines had an effort in inhibiting the dermatophytes,the propolis was the best,the range of MIC and MFC was 0.4692 mg/ml-0.93850 mg/ml and 0.9385 mg/ml-1.887 mg/ml.Results2:Under the orthogonal-design method combined with pharmacodynamics,the optimal proportion of five ingredients(the ethanol extract of radix sophorae flavescents,cortex dictamni,Stellera chamaejasme L,Cortex Pseudolaricis)in the "KXPL" was selected.The orthogonal-design table L16(4 5)was used and the ratio of inhibitory fungi was used as index.Under mathermatical statistics method,the optimal concentration of "KXPL" for 24h 32h 40h 48h were 6.0375 mg/ml、4.067mg/ml、4.5375mg/ml、7.5 mg/ml,3.7543 mg/ml;6.0375 mg/ml、4.067mg/ml、4.5375mg/ml、7.5 mg/ml,3.7543 mg/ml;6.0375 mg/ml、4.067mg/ml、2.2687mg/ml、3.75 mg/ml 0.4662mg/ml;12.075 mg/ml、4.067mg/ml、2.2687mg/ml、7.5mg/ml,0.4662mg/ml.radix sophorae flavescents played an important role in the inhibiting the Dermatophytes.Propolis can improve the effect of inhibiting the dermatophytes for radix sophorae flavescents,cortex dictamni,Stellera chamaejasme L,Cortex Pseudolaricis.Results3:Established dermatophytosis animal model by Trichophyton mentagrophytes to observe 4 groups Compound traditional Chinese medicine by Pathogens in vitro separation,clinical pathology score and histopathological observation.Rabbit clinical symptoms after 4 days with four groups of compound Chinese medicine got good improve,clinical pathology scores for the Chinese traditional medicine were significantly lower than T5 group after 14 days(P≤0.05),the scores among each Chinese traditional medicine was not significant difference(P≥0.05)T4 group effect is best in four groups of compound traditional Chinese medicine(TCM).The lesion got the basic recovery,there weren’t mycelia and conidia in the corium layer by the PAS staining.Results4:The compound antifungal reagent was daubed on the naked skin of rabbits with 2 times a day.each time for 15days,the stimulation reaction of CAR to health skin and damaged skin were observed.Kun Ming mice then were orally administrated with CAR in order to observe toxic reaction,each time for 7 days.Results showed that compound antifungal reagents had no skin stimulation or avimlence.It is safe for skin use.
