Chrysanthemum morifolium is a traditional famous flowers of our country.This study on the one hand,with short-day sensitive chrysanthemum varieties 'zijiao' as the test materials,using real-time quantitative fluorescence PCR technology to study the expression characteristics of the circadian clock genes CmPRR.On the other hand,short-day sensitive chrysanthemum varieties 'zijiao' and short-day not sensitive chrysanthemum varieties 'aoyunbaixue' as the research object,compare two different chrysanthemum varieties in the flower bud diflferentiation process,process and the difference of gene expression three aspects,the results are as follows:1.CmPRR2 gene was cloned from chrysanthemum'aoyunbaixue' that open reading frame have 1605 bp,encoded 535 amino acids.CmPRR73 gene open reading frame have 1941 bp,encoded 646 amino acids.2.Space-time expression characterization results:In different development stages of 'zijiao',the expression pattern of CmPRR gene was CmPRR2,CmPRR73,CmPRR37,CmPRR7.3.Tissue specificity results:CmPRR gene expressed higher in the flower and stem of 'zijiao'.4.Photoperiod expression pattern:The expression order of the day was CmPRR2/CmPRR73?CmPRR7-CmPRR37.5.The pattern in continuous light condition:After 'zijiao' from long-day condition transfer to continuous light condition,CmPRR2 and CmPRR7 maintain the original pattern of circadian oscillation.After 'zijiao' from short-day condition transfer to continuous light condition,CmPRR2 and CmPRR37 maintain the original pattern of circadian oscillation.6.The pattern in continuous dark condition:After 'zijiao' from long-day condition transfer to continuous light condition,CmPRR7,CmPRR37 and CmPRR73 maintain the original pattern of circadian oscillation.After 'zijiao' from short-day condition transfer to continuous light condition,four PRR genes are maintain circadian oscillation pattern.7.'zijiao' expression pattern after night break:After night break,CmPRR2,CmPRR37 and CmPRR73 has similar expression trend to short-day,but during the period of night break appeared opposite pattern.8.Short-day sensitive chrysanthemum varieties 'zijiao' and short-day not sensitive chrysanthemum varieties 'aoyunbaixue' flower bud differentiation comparison results:(1)Time that 'zijiao' needed to complete the flower bud differentiation is more 13 days than 'aoyunbaixue',flower flower bud points starting period later 38 days than 'aoyunbaixue'.(2)'aoyunbaixue' and 'zijiao' starts acceleration after the budding flower bud differentiation.(3)Different flower bud differentiation period,order is CmPRR73?CmPRR37?CmPRR7.CMGI1,CmGI3,CMCOL1,CmCOL5,CmCOL13,CmCOL15 are flowering promoting factors.CmCOL14,CmCOL20,CmFT are flowering inhibiting factor.CmCOL9 gene effect is not obvious in the chrysanthemum flower bud differentiation.
