Cloning And Expression Of CONSTANS-like Controlling The Flowering Time Of Chrysanthemum That Not Sensitive To Light

Chrysanthemum is China’s traditional name,the world’s significant four fresh cut flowers.Now chrysanthemum is planted mostly in autumn,the time for supply are limited,so the value is reduced.Because chrysanthemum is short-day plant,the light cycle is the major factor of flowering,to cultivate a new variety that not sensitive to light cycle is good for technology production.A gene related to flowering time excavation is the precondition of realization of molecular breeding.The cloning of CO,which related to flowering time from Chrysanthemum that not sensitive to light,then switch CO to Chrysanthemum,it could get a new variety that not sensitive to light.In this study,The genes of CONSTANS-like (CmCOL1), which related to flowering time were isolated from Chrysanthemum morflorium ’aoyunbaixue’ by PCR and rapid amplification of cDNA ends (RACE),CmCOL1encoded383amino acids.CmCOL1protein had typical structures,including B-box1, B-box2and CCT domain.RT-PCR and qPCR indicated that CmCOL1expression were different in various stages and organs.When ’aoyunbaixue’ was at the stage of involucre primordial diferentiation,the expression of CmCOL1in the leaves was the highest,then in stem,then in flower,last in root.When compared CmCOLl exprssion in flower of different stages,we could find that the expression of CmCOL1at the stage of posterior petal was the highest,then involucre primordial,then the initial stage of floret primordial,initial petal last. When compared CmCOL1expression in the stem of different stages,it showed that the expression of CmCOLl at the stage of posterior petal was the highest,and a little at other stages.QPCR showed that,under short days,CmCOLl expression lowed first then went up later.It reduced in illumination stage,while rising in dark stage.Under long days, CmCOLl expressed little,it lowed first then went up later in illumination stage,and in dark stage it was opposite.CmCOL1gene had7copies through Southern hybridization.The results of the study shows that CmCOLl gene is a gene family and it was under control of circadian clock,the most important is that CmCOLl gene is one of regulation plant flowering time important genes.
