| We determined the preparation process of the Yinhuang Erchen mixtures.We established a method for determination of baicalin in the Yinhuang Erchen mixture,while the methods were established for identifying Baical Skullcap Root,Honeysuckle Bud and Flower,Liquorice Root and Dried Tangerine Peel.The prescription of Yinhuang Erchen Mixtures includes Baical Skullcap Root,Honeysuckle Bud and Flower,Ginger Processd Pinellia,Liquorice Root,Indian Bread and Dried Tangerine Peel.We used the orthogonal experiments to select the best extraction process parameters of the Yinhuang Erchen Mixtures.We based on Rate of dryness and Content of Baicalin in order to the evaluation factors of water addition,recovery time and recovery times,etc.The HPLC method was determined for the determination of baicalin in Yinhuang Erchen Mixtures by systematic applicability test,negative interference test,linear relation test,precision test,repeatability test,solution stability test,sample recovery test,durability test and content limit test.The layer chromatography was used to identify Baical Skullcap Root,Honeysuckle Bud and Flower,Liquorice Rootin and Dried Tangerine Peel.The optimum extraction process of Yinhuang Erchen Mixtures: We added to 8 folds water to Honeysuckle Bud and Flower,Ginger Processd Pinellia,Liquorice Root,Indian Bread and Dried Tangerine Peel to boil,then acceded to Baical Skullcap Root.This process was repeated 2 times,2 hours each time.The linear range of Baicalin was 0.04~0.40 mg/mL(0.2~2.0 μg),y=20000000x + 9669,r = 0.99995.The average recovery rate of Baicalin was100.1%,RSD was 1.06%,the product content limits was not less than 7.0mg/mL Baicalin.The conditions of layer chromatography were determined for Baical Skullcap Root,Honeysuckle Bud and Flower,Liquorice Rootin and Dried Tangerine Peel.The results were showed that have the good separation effects and few disturbance factor in the study.Conclusions: The production process of Yinhuang Erchen Mixture was determined,which is stable,reliable,easy to operate and low cost.A high performance of liquid chromatography method was developed for the determination of baicalin in Yinhuang Erchen Mixtures.The method has the characteristics of high sensitivity,precision and easy operation.The layer chromatography was exclusive,easy and quick for the determination of Baical Skullcap Root,Honeysuckle Bud and Flower,Liquorice Rootin and Dried Tangerine Peel. |
