Raddix Scutellariae named also gold dishes、camellia roots and rotten heart grass,which widely used in the pharmaceutical industry and light industry in China andother Southeast Asian countries raw material, has antibacterial、 anti-viral、anti-inflammatory、anti-allergic、antioxidant and anti-tumor functions. The mainactive ingredient of Raddix Scutellariae is Baicalin. Extraction and separationpurification methods of Baicalin in Raddix Scutellariae were studied, and the contentof baicalin in different parts of the Raddix Scutellariae was measured in this paper.The effect of the cμlture conditions on the production of baicalin was studied duringcell suspension cμlture using callus of Raddix Scutellariae root cμltured ingreenhouse as material.1. It was determined that the best method of baicalin extraction from RaddixScutellariae was microwave extraction among five extract methods which wereSoaking method, boiling method, alkali soaking method, μltrasonic and microwavemethod; The orthogonal test was designed to optimize the conditions of microwaveextraction. The resμlts showed that the optimum conditions of microwave extractionwere as follows:55%ethanol concentration was55%, extraction time was2h,Solid-liquid ratio was1:16, microwave time was80s, and the yield of extraction was5.196%.2. The conditions of baicalin adsorption chromatography purification werestudied, and the resμlts showed that the optimum conditions were as follows: bedvolume was2.6×60; the fluid speed was0.6mL/min. column temperature was25℃,the injection volume was10μl, and baicalin content in purified samples was85.3%.3. The content of baicalin in different parts of the Raddix Scutellariae wasmeasured. The resμlts showed that the baicalin content of Raddix Scutellariae rootwas most, that of Raddix Scutellariae stems and leaves was more, and that of calluswas least.4. The effect of the cμlture conditions on the production of baicalin was studiedduring cell suspension cμlture using callus of Raddix Scutellariae root cμltured ingreenhouse as material. The resμlt was showed that:0.5g/L concentration oflactalbumin hydrolyzate has promoting role on the flavonoids generation duringCallus cell suspension cμlture;0.01g/L concentration of yeast extract has promotingrole on the flavonoids generation when Callus cell suspension was cμltured for10days;1g/L concentration of YE has promoting role on the flavonoids release when Callus cell suspension was cμltured for15days;5mg/L sμlfosalicylic acid haspromoting role on the flavonoids generation when Callus cell suspension was cμlturedfor15days.0.1mg/LSA and5mg/L has promoting role on the flavonoids releasewhen Callus cell suspension was cμltured for10days.
