Lead is a toxic heavy metal applied widely in industrial production.Excessive lead can pollute soil and water.Lead in the environment can accumulate in human body and animal body,thereby harming them.Selenium,as a nutrient trace element(essential for metabolism of human and animal bodies),can alleviate the toxicity of lead.Therefore,the toxicity of lead on chicken testes is mainly analyzed in the paper,and and the alleviating effect of selenium on lead toxicity is studied in series.1-day-old male Hailan chickens were pre-reared for one week.60 healthy chickens were selected,which were divided into control group,selenium group,lead group and lead-selenium group randomly with 15 chickens in each group.The control group was fed with commercial diet and drinking water containing selenium of 0.49 mg/kg;the selenium group was fed with commercial diet and drinking water containing sodium selenite of 1 mg/kg;the lead group was fed with commercial diet and drinking water containing lead acetate of 350 mg/L;the lead-selenium group was fed with commercial diet the same as selenium group and drinking water the same as lead group.All chickens were euthanized,and the testicles were removed on the 90 th day of the experiment.The microstructure and ultrastructure of chicken testes were observed.The apoptosis of chicken testes was detected.The mRNA expression of endoplasmic reticulum stressrelated genes(GRP78,PERK,eIF2?,ATF4,CHOP)and apoptosis-related genes(caspase-12 and caspase-3)was detected.The results showed that:(1)The tissue structure of chicken testes was changed by lead poisoning,and selenium was capable of relieving chicken testicular injury caused by excessive lead accumulation.(3)Excessive lead accumulation induced GRP78,PERK,ATF4,eIF2 alpha and CHOP in chicken testes,thereby causing endoplasmic reticulum stress,and selenium relieved endoplasmic reticulum stress induced by lead poisoning(3)Lead poisoning induced caspase-12 and caspase-3mRNA expression in chicken testes,thereby resulting in apoptosis,and selenium relieved the changes of apoptosis-related gene expression induced by lead poisoning.The conclusions are shown as follows:(1)Lead poisoning can cause chicken testicular injury,and selenium can relieve testicular injury of chickens caused by lead positioning.(2)Lead poisoning can lead to endoplasmic reticulum stress of chicken testes,and selenium can relieve endoplasmic reticulum stress of chicken testes caused by lead poisoning.(3)Lead poisoning can induce apoptosis of chicken testes.Selenium can relieve apoptosis induced by lead poisoning.(4)CHOP/caspase-3 signal pathway involves in relieving effect of selenium on endoplasmic reticulum stress and apoptosis in chicken testes caused by lead poisoning.
