| China is the world’s largest producer and consumer of vegetables,and various vegetables occupy an important position in national life and economy.Verticillium dahliae is a soil borne vascular disease.Verticillium dahliae has the characteristics of strong virulence,wide host range and long survival time,so the prevention of verticillium wilt has always been a major problem in vegetables production.In 1998,Indian scientist Verma et al.first discovered and named Piriformospora indica,existed studies have shown that Piriformospora indica plays a significant role in plant growth and resistance to a variety of plant diseases.Currently,popular study of verticillium wilt,mainly on the of cotton verticillium wilt,so in this test we will shoose cotton as the main research material,around Piriformospora indica raise resistance of plants to verticillium wilt mechanism study,in order to get a set of relatively perfect system and apply to the production of all kinds of vegetables,to alleviate the disease happening of all kinds of vegetables verticillium wilt.In this study,the results of the plate confrontation experiment showed that the Piriformospora indica could inhibit the growth of Verticillium dahliae in vitro.The experimental results showed that the frequency and degree of verticillium wilt were significantly decreased in the cotton inoculated with Piriformospora indica.The results of i TRAQ experiment on total protein of cotton root system showed that among all 1069 identified proteins,there were significant differences in the protein expression of cotton root.Histological analysis was carried out on 104 trusted differentially expressed proteins.The results showed that 104 proteins were enriched to 38 GO entries.KEGG results showed that 104 differentially expressed proteins were enriched in 64 metabolic pathways.In order to further verify the expression of genes,we chose 4 genes inα-linolenic acid metabolic pathways and 4 genes associated with disease resistance function to do q RT-PCR detection,found that seven genes expressed in transcription level and protein level is consistent,but the expression of obvious difference,that may be because the protein modification after translation.These above results showed that Piriformospora indica played a significant role in enhancing the resistance to verticillium wilt in cotton.The colonization of piriform spores from India can affect the metabolic pathway of plant α-linolenic acid and thereby affect the biosynthesis of JA,as well as the expression of some disease-resistant genes.On the whole,plant disease resistance is a complex process,and the mechanism of Piriformospora indica could increase cotton resistance to verticillium wilt remains to be further explored. |
