China's annual total meat production is 85.468 million tons,producing 4 million tons of livestock and poultry blood.The total output of dried fruits is 160,000 tons,of which about 30,000 tons are defective.The vast majority of livestock and poultry blood and defective products are not directly developed into the natural world,resulting in large economic losses and serious environmental pollution.Therefore,the comprehensive development and utilization of defective and livestock blood has extremely important reality.significance.Our previous experimental study found that adding the defective products to the feed to feed the Hailan brown chicken can greatly improve the quality of chicken and eggs,especially the content of vitamin E and ?-carotene in the eggs produced has been significantly improved..In order to further explore the effects of alfalfa-containing feed on the antioxidant activity of livestock and poultry products,different concentrations of alfalfa feed were prepared to feed Hailan brown chicken,and the changes of weight gain,egg production rate,egg nutrient composition and chicken plasma antioxidant performance were investigated.The chicken plasma antioxidant peptide was further isolated and purified by screening and optimizing the enzymatic hydrolysis of plasma protein;then the mouse D-galactose oxidative damage model was established,and the antioxidant activity of the antioxidant peptide in the body was analyzed to provide biochemical products for development.Theoretical support and technical support.The experimental results are as follows:1.The concentration of radon in Hailan brown chicken feed was positively correlated with chicken weight gain,egg production and plasma protein content.Moreover,alfalfa feed has a positive effect on egg quality,and can significantly increase vitamin A,vitamin E and cholesterol levels,but has little effect on Huff units,protein height,egg yolk height and egg yolk width.2.Alkaline protease enzymatic hydrolysis of Hailan brown chicken plasma protein has the highest degree of hydrolysis,and has a significant effect on the antioxidant activity of chicken plasma protein hydrolysate.The superoxide anion radical scavenging rate of alkaline protease and trypsin hydrolysate was 76.63%±0.57% and 76.67%±1.17%,respectively;the alkaline protease hydrolysate had the largest reduction ability of 0.82±0.04;alkaline protease hydrolysis The liquid DPPH has the strongest ability to remove DPPH,which is 75.70%±0.29%.The alkaline protease hydrolysate has the best chelation ability for metal ions,which is 83.43%±1.06%.3.Optimized the process conditions of plasma proteolysis of Hailan brown chicken.The optimum conditions of alkaline protease enzymatic extraction were as follows: enzymatic hydrolysis temperature 45 °C,enzymatic hydrolysis time 6 h,enzyme to substrate ratio 6000 U/g,p H 7.5,the best hydrolysis degree,the result was 19.45±0.05 %.4.The mouse D-galactose oxidative damage model was successfully established.Plasma protein antioxidant peptide can effectively reduce the MDA content in mouse serum,reduce protein peroxidation,enhance the activity of antioxidant enzymes in mouse serum,and increase the role of antioxidant glutathione in three experimental groups.The best results were obtained in the middle dose group.HE staining of mouse liver confirmed that the model control group had obvious hepatic lobular lesions,which proved that superoxide had a greater liver damage.The other groups of liver lobular tissue structure is normal,which proves that vitamin C and antioxidant peptides have a strong protective effect on the liver.
