Xinjiang brown cattle(XBC)and its hybrid are the main breeds for beef production in Xinjiang.However,its molecular mechanisms and development of fat metabolism related meat quantity traits are lack of clear invesitigation in comparison with those of excellent domestic and foreign breeds.In the present study the three month-old weaned calves of XBC and Angus(ANG)were chosen,the physiological and biochemical indexes,the serum fat metabolic controlling hormones(i.e.,fatty acid syntheses,fatty acid-binding protein 4,hormone-sensitive lipase,lipoprotein lipase and leptin),the carcass traits,and the expressions of fat metabolism regulating related gene's m RNA(i.e.,FASN,FABP4,HSL,LPL and Leptin)and their product proteins were analized and compared between the two breeds.The aim of the study was to have a better understanding of the molecular forming mechanisms of the fat metabolicm related meat quantity traits and their development from the “molecule to interity” in XBC compared to ANG.The results were as followings:1.The white blood cells in XBC was extremely lower than that in Angus(P<0.01),whereas the hemoglobin was significantly higher than that of Angus(P<0.05).The serum phosphorus in XBC was extremely higher than that in ANG(P<0.01),the blood glucose in XBC was significantly lower than that of ANG(P<0.05).The serum leptin and HSL levels in XBC were significantly higher than those in ANG(P <0.05),and the FASN,FABP4 and LPL were etremely higher than that of ANG(P <0.01);the eye muscle area in XBC was significantly bigger than that in ANG(P<0.05),the intercostal fat thickness in XBC was significantly thiner than that in ANG(P<0.05),the meat/bone radio in XBC was extremely significant lower than ANG(P<0.01).2.The adipocyte area of the pericardial,perirenal and omental adipose tissues in XBC was significantly smaller than that in ANG(P<0.05),whereras the adipocyte area in the subcutaneous adipose tissue was not different from that in Angus(P>0.05).The adipocyte number per unite of the omental adipose tissue in XBC was significantly lesser than that of ANG(P<0.05).3.The m RNA expression of the pericardial adipose FASN,Leptin and LPL in XBC was significantly higher than that in ANG(P<0.05),the m RNA expression of the perirenal adipose HSL,LPL,FABP4 and FASN in XBC was extremely higher than that in ANG(P<0.01).There were no significant differences in m RNA expression of the omental and subcutaneous fat between two breeds.The protein expression of the pericardial,perirenal adipose LPL in XBC was significant lesser than that in ANG(P<0.05),whereras the protein expression of the FASN and HSL was not different between two breeds.The above results indicated that: Xinjiang brown cattle blood physiology & biochemistry,serum leveal of FASN,FABP4,HSL,LPL,Leptin hormone and partial slaughtering traits and adipose tissue cell area has a difference compared with Angus.Leptin?FASN?FABP4?HSL?LPL gene m RNA expression difference appear pericardial fat,perirenal fat adipose.These differences provide molecular reference for early fat targeting deposition of the Xinjiang brown cattle.
