| Actinidia chinensis is a unique Actinidia plant which favored by people.Actinidia chinensis‘Yangshi Jinhong 50’is a fine variety with great potentiality from Actinidia chinensis.Advantages of tissue culture seedling include short cycle,no seasonal limit,stable genetic trait and so on.In this study,explants in vitro culture were selected by stem with buds,leaves,petioles from Actinidia chinensis‘Yangshi Jinhong 50’,established a rapid propagation system and provided technical support for industrial seedling.In this study,leaves,petioles from wastes of transplanting.The test results are as follows:1.The best disinfection method of stem with buds:the stem with buds were treated with 75%alcohol for 30 s+0.1%mercuric chloride for 8 min,contamination rate was20%,browning rate was 26.67%,survival rate was 53.33%.2.The best medium used for producing axillary buds from stem with buds was MS+4 mg/L 6-BA+0.1 mg/L NAA,average germination percentage was 94.44%.3.The best medium used for proliferation from buds was MS+4 mg/L 6-BA+0.5mg/L NAA,average proliferation coefficient was 5.16,elongation range of stem was2.01~3.77 cm.4.The best medium used for directly producing adventitious buds of leaves was MS+2 mg/L 6-BA+0.2 mg/L NAA,average browning rate was 7.5%,average regeneration percentage was 92.5%;The best medium used for directly producing adventitious buds of petioles was MS+2 mg/L 6-BA+0.3 mg/L NAA,average regeneration percentage was 89.17%.5.The best medium used for rooting culture was 1/2MS+0.9 mg/L IBA,average callus diameter was 1.03 cm,average rooting rate was 100%,average root length was3.19 cm,average root number was 12.58.6.The best hardening-seedling method was opening lid in culture room for 7 d,sand transplanting survival rate of tissue culture seedlings of was 100%at present.The best sand transplanting method:indoor cultured for 2 weeks,outdoor cultured for 2 weeks,and observed change of seedlings root.The best transplanting medium was humus soil,transplanting survival rate was 100%.7.According to test data,this test constructed a tissue culture seedling factory of Actinidia chinensis‘Yangshi Jinhong 50’,and analyzed the economic benefit.The factory included tissue room of 50 m~2,test preparation room of 50 m~2,inoculation room of 20 m~2,transplanting greenhouse of 1300 m~2.The factory can produce about 191520 seedlings of Actinidia chinensis‘Yangshi Jinhong 50’per year,with sales reaching about 957600 yuan.Powerhouse cost was about 11000 yuan,depreciation charges of fixed assets was about16758 yuan,medicine cost was about 47498 yuan,water and power cost was about148203 yuan,labor cost was about 145000 yuan,totally came to about 368459 yuan.The average cost of per seedling was about 1.9 yuan.In cost,water and power cost accounted for 40.22%,labor cost accounted for 39.35%,medicine cost accounted for 12.89%.This cost accounting provided reference for study of reducing cost.The annual profit of the factory is about 589141 yuan. |
