The research maked Disporum cantoniense( Lour.) Merr.as the object and explores tissue culture seedling’s production and acclimatization technology of Disporum cantoniense( Lour.) Merr.. This paper reached the effect of adding exogenous hormones(6-BA, NAA, KT and IBA, etc.), sucrose and banana with different content to the breeding, sound seedling and rooting of tissue culture seedling,as well as explored the formula of most suitable culture medium; also reached the effect of opening cap and acclimatization’s time, substrate, temperature and light intensity to survival and growth of tissue culture seedling, to screen the best conditions of acclimatization of tissue culture seedling. Through the research to best medium formula and suitable acclimatization conditions to establish tissue culture seedling production and transplanting technology of Disporum cantoniense( Lour.)Merr., to lay a foundation for factory production in the future.The main results are as follows:1. with 1 node stem of Disporum cantoniense( Lour.) Merr as the explants of this experiment, the best culture medium was MS+6-BA 1.2 mg/L + KT 0.45 mg/L+ sucrose 20 g/L, whose effect of inducement of overgrown bud is optimum and sprout quantity was the most, when the multiplication coefficient was 6.79,the number of buds reached 160.67,and the shoot height run up to 6.56 cm.2.The content of banana in culture medium significantly influenced the increase of the effective seedling rate(height is greater than or equal to 1 cm) and the stem diameter, while the content of sucrose could obviously worked on the increase of shoot height. Relatively better sound seedling culture medium was MS+ 6-BA 1 mg/L+ NAA 0.2 mg/L+ sucrose 30 g/L + banana 50 g/L,when the effective seedling rate was 43.77%, the shoot height reached 1.32 cm,and the stem diameter was only 1.27 mm.3. The different concentration of NAA directly affect the growth status of seedlings, it also had certain effect on the increase of rooting rate and improve the condition of rooting.While the different concentration of IBA had significant effect on root number, root length, and there was great influence on rooting rate, root number and root diameter in the medium with different content of banana. The optimum rooting medium was MS + AC 0.3 g/L +NAA0.5 mg/L+IBA0.5 mg/L+banana 75 g/L, under this condition,the rooting rate was 84.44%, the rooting number could achieve 9.39, the rooting length was only 0.69 cm,and the rooting diameter could reach 1.28 mm, moreover, the seedling was in good condition,its height increased 2.12 cm,and the stem diameter was 1.27 mm.4.It obviously effected the survival rate of tissue culture seedlings which to be opened the bottle to training seedling for different length of time, and the differences in the impact of the number of neonatal leaf was significantly, while there wasn’t significant effects on increment of seedling height. The best way to cope with them is that opening the bottle for 9 days, then the survival rate could be 94.45%, and the increment of seedling height and the number of neonatal leaves was 4.05 cm and 1.81 respectively.There was no significant differences in the effects on the survival rate and the increment of seedling height of Disporum cantoniense( Lour.) Merr. on different kinds of matrix, while the effect is obvious on the increasing number of new leaves.The most suitable medium of transplanting is using only vermiculite, when the survival rate could be 87.62%, the increment of seedling height was 3.68 cm, and the number of neonatal leaves could achieve 1.37.It would influence the survival rate and growth status(the increment of seedling height and the number of neonatal leaves) of Disporum cantoniense( Lour.) Merr.significantly in different temperature. The temperature was beneficial to survival at15~20℃, and 20℃~25℃ conducive to the growth of seedlings, while it would be dehydration or decay to dead when the temperature was too high.Effects of light intensity on the survival rate and growth status(the increment ofseedling height and the number of neonatal leaves) of Disporum cantoniense( Lour.)Merr. had no significant effect. While the survival rate and increment of seedling height and the number of neonatal leaves could reach the best states when the light was 6400 lx, they could achieve 83.33%, 3.82 cm and 2.39 respectively.
