The establishment of tomato efficient regeneration system is the premise and basis of tomato genetic engineering breeding,heterosis fixation and excellent germplasm preservation.In this experiment,tomato cotyledons and hypocotyls of six different genotypes including Zhongshu No.4,baiguoqiangfeng,Maofen 802,American Red General,Hong Shuai 909 and Micro tom were used as explants,and their callus,adventitious buds and adventitious roots growing situation were observed under different hormone concentrations ratio,the test results are as follows:1.In the disinfection treatment of tomato seeds,the germination rate of seeds was the highest with the concentration of 7% sodium hypochlorite for 10 minutes under different disinfection times,and the germination rate of Hongshuai 909 was highest at 93.33%.Besides that,different genotypes tomato seeds in hot water dip are germinated early.The germination rate of Hongshuai 909 seeds is up to 96.00%.2.Under the different concentrations of 6-BA+IAA,the adventitious bud induction rate of hypocotyls of different genotypes was higher than that of cotyledons.The addition of 6-BA alone does not effectively differentiate adventitious buds.In addition,the adventitious bud induction rate of Meiguohongjiangjun and Hongshuai 909 was higher than that of Zhongshu No.4,Baiguoqiangfeng and Maofen 802,even higher than the control material Micro tom,indicating that the adventitious bud induction effect of red fruit is better than that of pink fruit.3.By observing the induction of adventitious buds of cotyledons and hypocotyls of different genotypes at different concentrations of 6-BA+IAA,the adventitious bud rate of cotyledon of Hongshuai 909 at 6-BA 1.0 mg/L+IAA 0.5 mg/L hormone concentration was 88.50%.The highest rate of adventitious bud induction was 93.83% at 6-BA 1.0 mg/L+IAA 1.0 mg/L,which was higher than other tomato materials.4.Under the different concentration ratio of TDZ+IAA,the highest adventitious bud induction rate of hypocotyls was TDZ 1.0mg/L+IAA 0.2mg/L,the rate was 75.33%.It was also found that cotyledons and hypocotyls produce a lot of root hairs during the induction of callus and adventitious buds,approaching 100.00%.5.The optimum hormone concentration ratio of adventitious bud rooting of different genotypes of tomato was 1/2MS+IAA0.3mg/L,the root hair was thick,the number of rooting was large,rooting effect is better than other concentrations.Above all,the regeneration efficiency of Hongshuai 909 is significantly higher than that of the other five tomato varieties,and the high-efficiency regeneration system of Hongshuai 909 has been initially established.The regeneration system of other tomato varieties still needs further optimization.
