| Morangaceae oleifera Lam is a perennial tropical deciduous tree.At present,hot wood is mainly distributed in Yunnan,Hainan,Guangdong,Guangxi,Taiwan and other provinces and cities in the south of China.The existing acreage of spicy wood is about 5330hm2.At the present stage,it is the key to popularize and apply the excellent genetic quality of hot wood in large area to construct the artificial cultivation base of hot wood industrialization in our country,and to use plant tissue culture technology to carry out rapid propagation and seedling breeding.In this study,different culture medium and antimicrobial agent were selected for tissue culture test,in order to select suitable medium formula and the ratio of antimicrobial agents in each stage,and find out a better combination of technical parameters of tissue culture.Thus,an efficient tissue culture and rapid propagation system of spicy wood was constructed to alleviate the current situation of the large demand for hot wood seedlings,and to provide a scientific theoretical basis for the realization of large-scale production of spicy wood seedlings.The main conclusions are as follows:1.In the process of tissue culture and rapid propagation,the type and proportion of culture medium is the key,except for the condition of Mori.According to the results of the experiment:(1)the most suitable medium formula for the induction stage of tissue culture was MS+6-BA0.5mg/L+NAA0.04 mg/L.(2)MS+6-BA0.5 mg/L+NAA0.02 mg/L was the most suitable medium for tissue culture and proliferation.(3)the suitable medium for rooting was 1/2 MS+NAA0.2 mg/L+IBA0.3 mg/L.2.In the process of tissue culture,the effective prevention and control of pollution can improve the reproduction coefficient and the survival rate of transplanting,and reduce the production cost of seedlings.The results showed that the best disinfection effect was obtained by using 1g/L Aikeli in the explant disinfection stage.Compared with the blank control group,the contamination rate was lower by 283%,the germination rate was 32%higher,and the burn rate was 47%higher than that of the blank control group.At the induction stage,the storage rate and germination rate of Zhipeiling(0.08 g/L)were the highest,the average preservation rate and the average germination rate were 9%and 11%higher than those of the control group,respectively.Compared with the control(blank),it increased by 51%and 42%respectively.The pollution rate of 0.06 g/L and 0.08 g/L Zhipeiling decreased by 44%and 82%,respectively.0.06 g/L Zhipeiling had the best antipollution effect.The seedling growth effect of rooting culture with 0.06 g/L and 0.08 g/L Zhipeiling was better than that of Yipeilong.The seedling height was increased by 13%and 17%,the root length was increased by 6%and 11%,and the rooting rate was increased by 6%and 9%,respectively.The seedling height,root length and rooting rate were decreased by 16%,11%and 10%,respectively,at 0.03 g/L of Zhipeiling compared with the control group(blank),indicating that 0.03 g/L Zhipeiling had a significant effect on bacteriostasis at the same time.The root length and rooting rate can be increased. |
