Abstract/Summary:
Objective:To explore the different concentrations of sodium fluoride of mice MC3T3-E osteoblast cell morphology and effects on intracellular Ca2+,NO concentration and the level of ROS and mitochondrial membrane potential on apoptosis of osteoblasts.Methods:in vitro culture MC3T3-E1 osteoblast cell and conduct the fluorine experiment.sodium fluoride concentration,time gradient model,access MTT growth curve,determine the effect of fluoride on osteoblast proliferation;by DAPI staining and HE nuclear morphology and cell morphology;using a fluorescent probe Fluo-3 AM and DAF-FM DA dye labeled fluorine osteoblasts,by flow cytometry in osteoblasts Ca2+,NO concentration.The use of fluorescent probes DCFH-DA and JC-1 dye fluoro on labeled osteoblasts,were detected and analyzed by flow cytometry.Results:With increasing fluoride concentration,MTT growth curve changes in cell morphology more volatile,increasing the concentration of osteoblasts NO Ca2+,and for a long time will make the role of fluoride to reduce the activity of cell life,weakening or absence of physiological responses,thus affecting NO,Ca2+ concentration.Conclusion:High concentrations of fluorine can affect cell proliferation,cell morphology,and even induce apoptosis in this process osteoblasts Ca2+,the concentration of NO is increased.Fluoride at high concentrations can damage the structure of osteoblasts,resulting in osteoblasts mitochondrial membrane potential and ROS levels change,which causes apoptosis.
