| The quality of wheat depends largely on the type and content of gluten.Gluten protein is composed of gluten and gliadin.Gluten mainly affects the elasticity of gluten,and gliadin determines the malleability and viscosity of gluten.Both Dof and b ZIP are transcription factors that exist widely in plants and play important roles in plant growth,development,stress resistance and metabolism.Studies in maize showed that Dof and b ZIP were involved in the synthesis of gliadin.However,the regulation mechanism of these two transcription factors in wheat is not clear.Based on the published genome sequence of wheat,the two transcription factors family,Dof and b ZIP,were identified and analyzed by bioinformatics method,and the genes specifically expressed in wheat endosperm were isolated by using transcriptome data.Further,excellent mutation sites were identified by candidate gene association analysis through gene cloning,sequence alignment and quality data analysis.The results are as follows:1.A total of 45 Ta Dof and 183 Tab ZIP transcription factors were identified from the wheat genome database,and most of the Ta Dof genes were mapped on chromosomes by Blast comparison,among which 45 Ta Dofs were distributed on 17 chromosomes except 7A,6B,7B and 7D.Evolutionary analysis showed that Ta Dof protein sequence was consistent with Arabidopsis classification,and the protein motif structure of Ta Dof family was predicted by MEME and SWISS-MODEL database.The results showed that these motif structures were highly consistent and conservative.2.According to the transcriptome expression data of wheat,the tissue expression characteristics of two major family members were analyzed: wheat Dof transcription factor Ta Dof1 was specifically expressed in spike,Ta Dof7 was strongly expressed in stem and grain endosperm,while Ta Dof10 was specifically expressed in endosperm;wheat b ZIP transcription factor Tab ZIP8 and Tab ZIP 51 were tissue-specific in grains.3.WGCNA(weighted gene co-expression network)was constructed,and the co-expression relationship between Ta Dof10 and three types of wheat gliadin genes was found by analysis,which indicated that Ta Dof10 involved in regulating gliadin gene expression;Ta Dof7 and Tab ZIP8 were co-expressed in endosperm.These results indicated that there was indeed a gene interaction between Dof and b ZIP transcription factors in wheat grains.4.In order to excavate the excellent allelic variation site of Ta Dof7,Ta Dof7 A was cloned from Zhengmai 366,a strong gluten wheat cultivar.Compared with Chinese spring,there were 38 polymorphic loci.Based on SNP(Single nucleotide polymorphisms)differences,a functional marker DOF7A-1 was developed,which could amplify 226 bp fragment in Zhengmai 366,but had no target band in Chinese spring.DOF7A-1 marker was used to detect 146 different wheat materials,77 of which had target bands and 69 had no target bands.According to the quality data,The protein content reached a significant level(P<0.05);the water absorption rate,formation time,stabilization time and weakening degree reached very significant level(P < 0.01). |
