As is known to all,the nutritional value of beef is very high,and as the living conditions of modern people become better and better,they are more strict in choosing beef products.Beef cattle in the production of business often need to transport,whereas the stress response caused by transportation can cause varying degrees of damage to the cattle,the body weight、 immunity and production performance is affected to some extent.At present,there are many anti-stress methods at home and abroad,which can improve the animal’s ability to fight stress,thus reducing the loss.Therefore,this test by studying two kinds of additives of beef cattle’s blood before and after the stress for transportation and the influence of muscle tissue,which discuss two kinds of additives transport stress resistance of effect.27 fattening healthy Liaoyu white cattle with similarAge、body weight and body condition,were selected randomly and assigned to control group、treatment group 1 and treatment group 2,with nine replicates per group.The control group was fed with normal diet,Treatment group 1 and group 2 were added in the fodder anti-stress 1(main ingredients with VC,VE,etc.)and anti-stress 2(Fe,Zn,Cu,etc.),The amount of anti-stress No.1 and No.2 was 51.35 g/day · ox and 40 g/day · ox.After 10 days,road transport was carried out for 5 h at 50~60 km / h,20 ° C and a relative humidity of 51 %.The beef cattle were weighed before and after transportation,and the blood of beef cattle was collected and the serum was prepared.After transportation,the muscle tissue of beef cattle was collected.The blood routine and other indicators were detected by automatic blood cell counting instrument;The concentrations of ACTH、Cor.、IL-2 and other indicatorswere detected by ELISA;The mRNA and protein expression of HSP27、HSP70 and HSP90 were detected by qRT-PCR and Western Blot;The residual amount of VC、VE、Zn and Fe in muscle and serum were detected by ELISA.The test results are as follows:(1)After transportation,the weight of beef cattle was significantly lower than before transportation,There was no significant difference between treatment group and control group,treatment group 1 and treatment group 2 before and after transportation(P>0.05).And there was no significant difference between the weight loss percentage and carcass weight before transportation(P>0.05).(2)The number of WBC was higher than before transportation(control group:P<0.01;treatment group:P<0.05);The number of RBC、MCV、and HCT were lower than before transportation(RBC control group:P<0.05;HCTtreatment group : P<0.01);RDW: The number of control group after transportation was lower than before transportation(P<0.01),and the treatment group was higher than before transportation(P<0.05);The MCH、RBC、MCHC were lower than those of the three groups after transportation(MCHtreatment group:P<0.05;MCHC three group:P<0.01);The number of HGB was higher than that of the control group before transportation(P<0.01),and the treatment group was lower than before transportation(P<0.01);PLT、MPV and PDW were lower than before transportation(PLT three group:P<0.01;MPV control group:P<0.01;PDW control group:P<0.05);PCT was lower than the control group before transportation,and the number of treatment groups increased.(3)The Concentrations of ACTH 、 Cor.、 IL-2 、 IFN-γ and HSP70 increased after transportation than before transportation,the treatment group was significantly lower than the control group before and after transportation(P<0.05).The concentrations of TNF-α 、 IgA 、 IgG and IgM were significantly lower than those before transportation(P<0.01).The two treatment group was significantly higher than the control groups before and after transportation(TNF-α treatment group 2:P<0.01;IgA、IgM treatment group 1:P<0.01,treatment group 2:P<0.05;IgG treatment group 1:P<0.01,treatment group 2:P<0.01).(4)The relative expression levels of HSP27、HSP70 and HSP90 mRNA were significantly lower in the treatment group 1 than control group(P<0.05).The expression of HSP27 mRNA in treatment group 2 was significantly higher than treatment group 1(P<0.05).(5)The relative expression of HSP70 and HSP90 protein: The expression level of the treatment group was significantly lower than that of the control group(P<0.05).(6)The residual amounts of VC、VE、Fe and Zn in muscle tissue did not exceed the national standard.Conclusion: Through the experiment concluded,anti-stress additive by changing the number of beef cattle routine blood、serum concentration of hormones、cytokines、immune factor and the relative expression of stress proteins in the beef muscle tissue,to reduce the damage caused by transportation stress to beef cattle,anti stress of No.1 has better resistance to transport stress than anti stress of No.2,and to improve the theoretical basis for the promotion and application of anti-stress additives.
