This research is aimed to study the effect of transport stress on the levelsof hormones, cytokines and acute phase proteins (APPs), explore the relationshipbetween these indicators and transport stress, and provide data basis for further studyof using blood hormones and APPs as indicators of cattle health. This research usedXia-nan beef cattle as the experimental animal and simulated transport stresssyndrome of beef cattle (TSSBC). The results of this research is hoped to providescientific data and basis for the control of TSSBC.In the experiment,8-month300kg healthy xia-nan cattle (♂) were selectedrandomly for long-distance transport. The transport lasted for20h and no water andfood was provided during the entire transport. Respectively, before transport-1h,0h;transport2h,4h,7h,10h and20h;15h,30h,2d,3d,7d,15d and21d aftertransportation, blood samples were collected from jugular vein, and the lymphocytesand serum were separated. The mRNA transcript levels of serum amyloid A (SAA)and haptoglobin (HP) in Peripheral blood lymphocytes of transportation stress bovinewere detected by Real-time fluorescence quantitative RT-PCR technology; serumC-reactive protein (CRP) levels was Detected by ELISA; the levels ofhormoneadrenocorticotrop(h)ic hormone (ACTH), cortisol (Cor), interleukin-6(IL-6)and tumor necrosis factor-α (TNF-α) was analyzed by radioimmunoassay.SAA mRNA transcription level began to rise slowly at transportation4h, andcontinued to rise in transport then peaked at15h after transportation. The levelincreased significantly (P<0.01) at15h、30h after transportation in cattle peripheralblood lymphocytes, then the expression level returned to normal levels after3d. HPmRNA transcript levels rose slowly at transportation2h reached a peak attransportation6h, then slowly returned to normal level at30h after transportation.And significantly increased (P<0.01) at transportation6h,10h; The level of CRP roseslowly after the start of transport, reached the peak point at transportation20h andthen slowly returned to normal level at7d after transportation. Contrast with1hour before transportation, Cor increased significantly (P<0.01) at transportation10h、20h,15h、30h、2d、2d after transportation. Contrast with1hour before transportation,ACTH was increased significantly (P<0.01) at transportation2h、4h、10h、20h,15h after transportation, then return to normal level at30h after transportation. COR inthe whole transport process was significant higher (P<0.05), then slowly return tonormal level at arrived2d. IL-6increased significantly (P<0.05) at transportation4h,and was extremely significant higher(P<0.01) at15h after transportation, thenreturned to normal levels after2d. TNF-α was increased significantly (P<0.01) attransportation6h, then returned to normal level at arrived30h.The results showed that: SAA, HP, CRP, COR and ACTH may be used aseffective indicators of animal welfare detection under transport stress in cattle. ACTH,COR, IL-6and TNF-α increase under short time transport stress, however a long timeof transport will cause immune suppression.
