6-month years old,one years old,two years old of male and female Weining sheep and Guizhou semi-fine wool sheep as the research objects,the experiment used molecule methods to cloned CDS region of relation genes about JAK/STAT signal transduction(SOCS7 and GFAP),meanwhile to analysis biological information.In addition,the experiment used real-time q PCR method to analysis m RNA experssion rule of SOCS7 and GFAP in heart,liver,spleen,lungs,kidneys,small intestine and brain in different growth phases of different genders,to obtain m RNA experssion rule of SOCS7 and GFAP in Weining sheep and Guizhou semi-fine wool sheep.The result is as show:(1)The whole sequence of the CDS region of the SOCS7 gene of Weining sheep and Guizhou semi fine wool sheep was cloned successfully,and two different kinds of variable splice bodies were found,named SOCS7-V1 and SOCS7-V2,respectively.Further comparison of the sequences found that the difference between the length of the CDS region of the two variable splice bodies was caused by the retention of the fifth exon of the sheep gene fifth and the partial exon of the sixth exon during the splicing.The length of SOCS7-V1 gene CDS region is 1827 bp,encodes 609 amino acids,the length of CDS region of SOCS7-V2 gene is 1662 bp,encoding a total of 554 amino acids,and SOCS7-V2 lacks 165 bp base pairs and 55 amino acids compared to SOCS7-V1.Through further analysis,it was found that different alterable splice bodies resulted in the change of the isoelectric point,molecular mass,the two stage structure and the three structure of the protein.(2)The CDS region complete sequence of GFAP gene of Weining sheep and Guizhou semi fine wool sheep was successfully cloned in this experiment.Weining sheep and Guizhou semi fine wool sheep GFAP gene CDS region length is 1407 bp,encoding a total of 469 amino acids.(3)Through the exploration of the expression of two variable splice bodies of the SOCS7 gene of Weining sheep and Guizhou semi fine wool sheep on the level of m RNA,it is found that two variable splice bodies of the SOCS7 gene are expressed in different degrees in Weining sheep and Guizhou semi fine wool sheep,among them,the total expression of two alternative splicing variants of SOCS7 gene was almost the same in Weining sheep and Guizhou semi fine wool sheep.The highest relative expression was in the spleen,followed by lung,kidney,heart,liver and brain tissue.The expression of SOCS7 gene in Weining sheep and Guizhou semi fine wool sheep was significantly different in the same tissues at different sex and growth stages.And with the increase of the age of Weining sheep and Guizhou semi fine wool sheep,the relative expression of the variable splice body of SOCS7 gene showed a small increase in the tissue.In Weining sheep and Guizhou semi fine wool sheep,the expression of SOCS7 gene in spleen,heart,lung,liver and brain tissues was slightly lower than that of ewes in five tissues.(4)explore the expression of GFAP gene in Weining sheep and Guizhou semi fine wool sheep at m RNA level,GFAP gene is expressed in Weining sheep and Guizhou semi fine wool sheep to varying degrees,but the relative expression of GFAP gene in Weining sheep and Guizhou semi fine wool sheep is the highest in the brain tissue.
