Ginger(Zingiber officinale Rosc)is a dual-purpose crop with high economic value.The small yellow ginger from Liupanshui area in Guizhou Province is a cultivar that contains high level of ginger oil,fine fiber and has a good taste which make it has strong market competitiveness.Small yellow ginger is mainly multiplied by vegetative propagation,however,due to the repeatly reproduction the virus were accumulated in ginger,which leads to the degradation of ginger varieties and the deterioration of quality.Artificial seed technology can guarantee the genetic stability of the superior traits of cultivars,and its application in production can improve the yield and quality of small yellow ginger.In this study,the artificial seed technology were developed and established of small yellow ginger.The main results of this study were shoun as followed:(1)The Artificial Seed Production Technology of Small Yellow GingerThe stem tip cultivation and heat treatment technique were used to produce the viruss-free buds,which were used as artificial embryos of the artificial seed.The artificial seed coat were composed from 4.0% sodium alginate,2.0% chitosan and 2.0% calcium chloride.The artificial endosperm was made from MS,4.0 mg/L 6-BA,1.0 mg/L NAA,0.3%activated charcoal,0.1 mg/L penicillin,0.1% sodium benzoate and 2.0% sucrose.After 10 minutes of complexation the artificial seed of the small yellow ginger.The seed can be germinated in the aseptic environment,with the germination rate at 86.67% and seedling rate at 83.33%.(2)Storage and Germination of Artificial Seeds were obtained Small Yellow GingerThe storage and germination condition of small yellow ginger were studied,the results shown that the artificial seeds had the highest germination rate(63.33%)and seedling rate(60.00%)when stored at 4°C for 10 days.By sprayed the MS medium,6-BA and NAA(1mg/L)on the artificial seed and sowed the artificial seeds in the composite matrix(nutritional soil: vermiculite=2: 1),the germination rate and seedling rate of the artificial seeds were reach to 61.67% and 58.33% in the open environment.The results shown that with the proper storage and germination conditions the growth of artificial seeds can be promote.(3)Virus Detection of Tissue Culture Seedlings of Artificial SeedIn this study,enzyme-linked immuno sorbent assay(ELISA)was used to detect the TMV content of artificial seed tissue culture seedlings.The results showen that the TMV content in the control group was 3.336 ?g/L while the average value of TMV content was0.4863 ?g/L in the group of artificial seed tissue culture seedlings.The concentration of TMV which was obtained by artificial seed technology of small yellow ginger was obvious lower than the control group.Therefore,it indicates that grown from the artificial seed of small yellow ginger do not affected by TMV.
