Plant virus interacts with insect vectors during the process of plant virus transmisson.An area of deficient in the field is whether bacterial symbionts of insect vectors are involved in the virus transmisson process.Aphid is the transmisson vector of Cucumber mosaic virus(CMV).Previous research showed that,Myzus persicae(Sulzer)endosymbiont,Buchnera ahpidicola can affect the interaction of aphids and plants in the process of feeding and colonizing,and this effect may facilitate virus transmission.Our research aimed on this scientific question,and surveyed the effect of Buchnera on aphid performance,and effect of CMV infection on Buchnera of aphids,and effect of Buchnera on plant volatiles and further investigated the role of plant volatile in virus transmission.The main results are as follows:1.The expression of Buchnera dnaK in ahphids treated with 200 μg/mL rifampicin was significantly decreased.2.We compared the growth and reproduction of Myzus persicae untreated and treated with rifampicin on three kinds of vegetables.The results showed that after treated with 200 μg/mL rifampicin,the reproduction,life span and body weight of aphids were significantly lower than that of the healthy aphids.3.The expression of Buchnera dnaK in ahphids feeding of aphids on CMV-infected plants was significantly decreased.4.The aphids acquired CMV or treated by rifampin altered the component of the plant volatiles after feeding on plants,the relative content of repellent volatiles were increased and the relative content of attractive volatiles were decreased.5.The aphids acquired CMV and treated by 200μg/mL rifampin were preferred to healthy plants,and virus transmission was promoted.According to the results we can speculate that the reduction of Buchnera density altered the component of plant volatiles,leading to the increased content of repellent volatile and the decreased content of attractive volatile,thereby aphids changed their preference to healthy plants,and promote CMV spreaded.
