In recent years,the incidence of canine breast cancer has been increasing gradually,and has become one of veterinary clinical common tumor diseases.There is no ideal treatment plan,as the canine breast tumor mechanism is still unclear.By building Experimental animal models,we are able to simulate the occurrence and development of diseases which also shorten the time and cost o f research.In this study,GFP was labeled in canine breast tumor cell lines CHMp and CHMm,The tumor formation model of nude mice was established by using the cells mentioned before,and observing tumor dynamic evolution,occurrence and development regularity in order to explore its biological characteristics.After in-depth study of canine breast tumor treatment plan,we can provide an animal model for pathogenesis.This experiment used lentivirus importing GFP into canine breast tumor C HMm and C HMp cells,by using limited dilution analysis,we successfully selected canine breast tumor cells CHMm-GFPand CHMp-GFP which markedby green fluorescent protein.Furthermore,we used cell suspension injections to inject stable expression of GFP canine tumor cells CHMm-GFP and CHMp-GFP into nude mice’s vivo,and finally established nude mouse tumor model.After that,we observed the state of the nude mice,and recorded the nude mice body weight and the size of the mass nude mice respectively.We dissected the nude mice in the different period of time(25 days,35 days,45 and 60 days),and then using the detection of mice in vivo imaging,isolated organ imaging and pathology.By comparison of different cell lines’ biological characteristics in nude mice.Results:Limited dilutio n method of canine mammary gland tumour cells C HMm-GFP and CHMp-GFP green fluorescent protein expression rate is high.There is no change in the cell shape and state after multiple cell passage,and green fluorescence signal is stable.After the cell suspension was injected into the right side of the second pairs of papilla of nude mice,the lump was visible on the sixth day,The curve of lump growth showed that the volume growth rate of nude mice in the two groups was different after inoculation,but the mass of CHMp-GFP group increased rapidly after inoculation for 30 days.25 days after inoculation nude mice in vivo imaging,CHMm-GFP nude mice in vivo fluorescence signal is strongest,the CHMp-GFP is in 35 days,nude mice in vivo fluorescence signal is the strongest.Two groups of at least 25 days after inoculation in vitro organ imaging,each have 1 nude mouse lymph nodes only began to emerge fluorescence signal,within 35 days 3 nude mouse lymph nodes have shown fluorescent signal,other organs without fluorescent signal.Histopathology showed that two cell lines lead to subcutaneous mass were tumor,two types of cell into a tumor rate is 100%.25 days of pathological sections,two cell lines were 1 nude mice tumor metastasis to the lymph nodes,in 35 days,45 days and 60 days when dissected nude mice,tumors were found has metastasized to the lymph nodes.In this study,we successfully labeled GFP into CHMm and C HMp cells and established the nude mice tumor model to explore its biological characteristics,and found that CHMm-GFP tumor model in the early growth of faster but smaller tumor mean volume,CHMp-GFP tumor model in the late growth speed and large mean size of tumors,and tumor in two nude mice models can be transferred to the lymph nodes.This study provides basis for further research on canine breast tumor.
