Hybrid tomato varieties usually give high yield,good quality characteristics,disease resistance,and abiotic stress tolerance.Therefore,the development of excellent tomato hybrid varieties has become the current trend of tomato breeding.However,tomato hybrid seeds are usually produced by hand emasculation and pollination,which in turn further increases the price of the hybrid seeds and the risk of false hybrid seeds due to self-pollination.Excellent tomato male sterile line has the following characteristics:completely sterile,pollination can be done without emasculation or hand emasculation can be done easily and fast,no effect on seed yield after pollination.The use of excellent tomato male sterile lines for hybrid seed production,not only can reduce the cost of seed production,but also can improve the purity of hybrids.Therefore,the screening of male sterile lines,the mapping of male sterile genes and the creation of male sterile breeding materials have important research significance and application value.In this study,the phenotype of tomato male sterile line 2-461S?ms-31?was analyzed,and its application prospect was deduced.The ms-31 locus was preliminarily mapped and the around molecular marker was developed.The detail of these results are as follows:1)The appearance of flowers was observed.It was found that the male sterile line 2-461S?ms-31?flower was slightly smaller,stigma was exposed,and the anther was yellowishgreen.2)The pollen was detected with acetic acid magenta.The flowers of 2-461S?ms-31?werefound without pollen.3)The fruit number were investigated.It was found that 2-461S?ms-31?had a smallamount of fruit set and could not cause 100%sterility.4)MS-31 gene was mapped to a range of 364644-bp between SL2.5ch10:3211054 toSL2.5ch10:3575698 on chromosome 10 by using a F2 population that was derived fromthe cross between 2-461S and Solanum pimpinellifolium LA1589.According to theITAG2.40,the interval encode 21 putative gene.5)Transcriptional expression characteristics of these 21 genes were analyzed.Among themSolyc10g009280.2,Solyc10g009330.2,Solyc10g009350.2 and Solyc10g009400.2 werehighly expressed at level in young flower buds and anthesis flowers.
