The purpose of this study is to do the comparative demonstration of the comparative test on three levels(molecular,cellular and individuals)between new long-acting recombinant follicle stimulating hormone(FSH-CTP-vFc,rFSH)and pregnant mare serum gonadotropin hormone(PMSG)on the market was used to timed artificial insemination(TAI)of replacement gilts,through pharmacokinetic(DMPK)and B-ultrasound the development of drug-taken follicles,trying to use rFSH to instead PMSG for large-scale pig farms and resolving the insufficient problem and high cost problem of PMSG caused by seasonal and limited production.The experiment results as following:Did the intramuscular injection castrated young boar through the high-gradient,medium-gradient and low-gradient rFSH and PMSG.The DMPK results show that the half life period of rFSH is 55.28±15.75h and the half life period of PMSG is 44h.That is to say,the half life period of rFSH is longer than PMSG.The results of in vitro observation of follicular development of estrus replacement gilts by different models of B-ultrasound and follicle size measurement by real-time slaughter are as follows:the linear equation of type 1 ultrasonic B between measuring mean value X and real mean value Y is y=0.8149x+3.0792 and R2=0.8721.The linear equation of type 2 ultrasonic B between measuring mean value X and real mean value Y is y=1.0079x+0.9699,R2=0.8031.That their R~2 is bigger than 0.8 means that the linear dependence of measuring mean value X and real mean value Y is strong.The ultrasonic B can clearly imaging the follicles of estrus replacement gilts and the linear dependence of measuring mean value X and real mean value Y is strong,that is to say,in real production,we can accurately predict the follicles size and determine the period of ovulation through the ultrasonic B for replacement gilts.The replacement gilts were treated by rFSH and PMSG respectively,and the slaughter was carried out in three stages of estrus.We used rFSH in timed artificial insemination regularly,and the follicular maturation was longer than the group of PMSG,which was consistent with the DMPK results.In the production practice,the insemination time of using RFSH in timed artificial insemination regularly needs to be delayed to sure that the mismatch rate is reduced and conception rate is increased.The timed artificial insemination experiment using rFSH and PMSG for batch replacement gilts entering the breeding units resulted in the following:rFSH(1500U/head)can replace PMSG(1000 IU/head)for German timed artificial insemination.In the treatment of estrus-prone replacement gilts,the ratio of rFSH 2000U+hCG500IU has the same or superior results as PG600,and can be used in place of PG600.
