Mosso bamboo(Phyllostachys edulis)is widely used and plays a very important role in China’s forestry production.However,adverse conditions in the natural environment will bring different degrees of damage to Mosso bamboo.Accumulating evidence suggests that VQ motif-containing proteins in rice(Oryza sativa),arabidopsis(Arabidopsis thaliana),and maize(Zea mays)play fundamental roles in response to various biotic and abiotic stresses.However,few studies have been pubilished is known about the functions of VQ family proteins in Moso bamboo(Phyllostachys edulis).In this study,it start with bioinformatics,through gene structure,conservative motif,sequence alignment,phylogenetic tree and promoter regional analysis,etc.to analyze that whole genomic VQ protein family of moso bamboo,We downloaded expression profile data to explore Pe VQ gene expression levels in various tissues or developmental Stages.And then,we analyze the expression pattern of the VQ gene under the treatment of PEG,SA and ABA stress.Subsequently,the gene Pe VQ28 was cloned from the bamboo genome,and it was analyzed and verified by subcellular localization experiment,yeast two-hybrid experiment and Bi FC experiment.The main results obtained in this study are as follows:1、29 VQ family genes were identified from the bamboo genome and compared with the VQ family genes of Arabidopsis thaliana and rice.These genes were named(Pe VQ1~ Pe VQ29),and divided into seven subfamilies.2、Pe VQ28 is located in PH01007611 date on one gene,gene length is 1659 bp,coding region of 513 bp,the encoding of a length of 170 aa protein,the protein isoelectric point is 5.75,the relative molecular mass is 18873.68 Da.3、Subcellular localization showed that Pe VQ28 protein was expressed in the nucleus.4、The yeast two hybridization and Bi FC experiment showed that the VQ gene can interact with the WRKY transcription factor and form the complex in the nucleus.5、In 300 m M mannitol and 0.5 μM ABA of MS,germination of wild type and transgenic strains have been suppressed,and the germination rate and root length of transgenic strains was significantly lower than that of wild Arabidopsis.
