Haworthia cooperi var.pilfera M.B.Bayer is a soft-leaf succulent plant belonging to the 12th genus of Liliaceae.It is a representative flower species in the family Liliaceae.Using the in vitro culture method for the rapid propagation of the Haworthia cooperi var.pilfera M.B.Bayer,it is possible to obtain a good-quality,stable-looking product.In this study,leaves of Haworthia cooperi,Haworthia cooperivar.truncata,and Haworthia obtusa var.pilifera were used as test materials and Haworthia davidii was used as the test material.The inflorescence of Haworthia cooperivar and Haworthia cooperivar was used as a test material,to study the effects of different growth regulators on callus induction,adventitious bud induction and proliferation,and adventitious root induction in Haworthia cooperi var.pilfera M.B.Bayer,aiming to establish a complete system of Haworthia cooperi var.pilfera M.B.Bayer tissue culture.Potted plants were used to test and collect data on the five main ornamental traits of plant height,crown width,window length,window width,and window thickness of different cultivars.The growth rules of Haworthia cooperi var.pilfera M.B.Bayer were analyzed and provided for the cultivation of Haworthia cooperi var.pilfera M.B.Bayer commodity seedlings.Theoretical basis.In addition,cuttings of the lower leaves of different varieties of Haworthia cooperi var.pilfera M.B.Bayer tissue culture seedlings were studied for cutting propagation techniques,and the survival condition,rooting status and seedling establishment status of the leaves were analyzed,which provided references and references for the rapid propagation of Haworthia cooperivar.pilferaM.B.Bayer.The main findings are as follows:1?Establishment of Rapid Propagation system in Vitro of different varieties of Haworthia cooperi var.pilfera M.B.Bayer(1)Callus induction test of Haworthia cooperi var.pilfera M.B,Bayer,through the orthogonal analysis of 6-BA,NAA and ZT(KT or IBA),screened the best formula for the recovery rate of Haworthia obtusa var.pilifera leaves is MS+6-BAlmg/L+NAA0.1mg/L+KT 0.2mg/L,the induction rate was 85.56%;the best formula for the inflorescence rate of Haworthia davidii inflorescence was MS+6-BA 0.5mg/L+NAA 0.01mg/L+KT O.lmg/L,The induction rate was 82.22%;the best formula for the leaf healing rate of Haworthia cooperivar.truncata and Haworthia cooperi was MS+6-BA1mg/L+NAA 0.1mg/L,and the induction rates were 83.33%and 86.67%;The best formula of Haworthia cooperivar was MS+6-BA1 mg/L+NAA 0.2 mg/L+KT 0.5 mg/L,and the induction rate was 82.22%.(2)Bud induction and proliferation test of Haworthia cooperi var.pilfera M.B.Bayer,by setting different hormone combinations,determine 1/2 MS+NAA 0.01 mg/L+6-BA 0.3 mg/L+KT 0.2 mg/Las the best medium for bud induction and proliferation of Haworthia obtusa var.pilifera,1/2 MS+NAA 0.01 mg/L+6-BA 0.3 mg/L+KT 0.1 mg/L is the best bud induction and proliferation media formula for Haworthia davidii,1/2 MS+NAA 0.1 mg/L+6-BA 0.6 mg/L+TDZ 0.01 mg/L is the best bud induction and proliferation medium formula for Haworthia cooperivar.truncata,1/2 MS+NAA 0.01 mg/L +6-BA 0.5 mg/L+ZT 0.1 mg/L is the best bud induction and proliferation medium formula for Haworthia cooperi,1/2 MS+NAA 0.01 mg/L+6-BA 0.3 mg/L+ KT 0.1 mg/L was the best bud induction and proliferation media formulation for Haworthia cooperivar.The clustered buds were cultured on the selected differentiation medium and the propagation coefficient was 5-6 times.Some of the seedlings had the seedling formation morphology.(3)Haworthia cooperi var.pilfera M.B.Bayer tissue culture seedling rooting test,by comparing the effect of different hormones on the average rooting rate and rooting rate of Haworthia cooperivar,it was concluded that the optimal rooting medium formula of Haworthia cooperivar was 1/2 MS+IBA 0.2 mg/L,and the rooting rate of Haworthia obtusa var.pilifera was 95.56%,The rooting rate of Haworthia davidii was 94.45%,the rooting rate of the Haworthia cooperi was 81.27%,and the rooting rate of Haworthia cooperivar.truncata was 90%.The rooting rate of the Haworthia cooperivar is 85.56%.2.Different varieties of Haworthia cooperi var.pilfera M.B.Bayer leaf propagation test(1)Rapid Propagation of Haworthia cooperi var.pilfera M.B.Bayer Leaf cutting,The survival rate of five varieties of Haworthia cooperi var.pilfera M.B.Bayer was more than 50%.The survival rates of Haworthia cooperi and Haworthia cooperivar.truncata were 80%and 73%respectively.The days of rooting and germination were 14 days and 15 days respectively,and the days of germination were 28 days and 30 days respectively.The survival rate of Haworthia cooperivar and Haworthia obtusa var.pilifera were 43%,47%,the days of rooting were 18 days,and the days of germination were 44 days and 49 days respectively.The days of Haworthia davidii rooting and germination were the longest,22 days and 54 days respectively,and the survival rate was 53%.(2)Plant height and crown width of five varieties of Haworthia cooperi var.pilfera M.B.Bayer.The plant heights were Haworthia cooperivar>Haworthia davidii>Haworthia obtusa var.pilifera>Haworthia cooperi>Haworthia cooperivar.Truncata,The crown width were Haworthia obtusa var.pilifera>Haworthia davidii>Haworthia cooperivar.Truncata>Haworthia cooperi>Haworthia cooperivar.3.Growth development analysis of Haworthia cooperi var.pilfera M.B.Bayertissue culture seedlingsFive varieties of Haworthia cooperi var.pilfera M.B.Bayer tissue culture seedlings were used as test materials.The growth and development indexes of the plant height,crown width,window width,window length and window thickness were measured every 60 days.Plant height of 60 days increase between 0.17cm-0.24cm,Crown width of 60 days increases between 0.13cm-0.24cm,Window length of 60 days increases between 0.70-0.90mm,and window width of 60 days increase between 0.35-0.84mm,Window thickness of 60 days increased between 0.07-0.20mm.
