Pinus serotina is originated in the southeastern coastal plains of United States,China began to introduce in1960 s.It have excellent features: strong adaptability,salt tolerance,cold resistance,fast growth,high biomass,germination ability,Sprouting ability,resistant to pests and so on.Because Pinus serotina's nuts are perennial persistent tree,the scales is not crack,resulting the natural regeneration slowly;But Pinus serotina's sprouting ability is very strong,so through the means of asexual propagation to accelerate the cultivation of Pinus serotina.This paper through tissue culture techniques and cutting techniques to study the asexual reproduction of Pinus serotina.(1)Key techniques of tissue culture of Pinus serotinaChoose the annual germination of newborn seedlings and seed germination buds for the explants,studying the disinfection of explants,induction and proliferation of axillary buds,seedling and rooting.Through direct organogenesis to establish a rapid propagation system of tissue culture.The main conclusions are as follows:Pinus serotina sterilization test of stem segments,the best is to sterilize 10 min in the middle of spring stem,and the pollution rate was only 0.8%.The best disinfection time of the seeds with 0.1% HgCl2 was 4 min and the pollution rate was 2.4%.The best medium for stem axillary bud induction was MS medium + 2.5mg/L 6-BA? 0.2 mg/L NAA,The differentiation coefficient of buds was 2.27.The best medium and hormone concentration ratio induced by axillary buds of seed buds were: MS medium +0.7mg/L NAA +1.0 mg/L 6-BA.The best medium for seedling culture is MS medium +0.5 g/L AC +0.4mg/L NAA,growth of adventitious buds was 1.1cm.The best medium root-induced is 1/2GD medium + 30 g / L sucrose,the root induction rate was 30.7%.(2)the key technology of cutting propagation on Pinus serotinaChoose the annual shoots of the annual growth of seedlings for the spike,study the effect of cuttings on rooting of about different cuttings surroundings ? different cutting type?different cuttings,and GGR different concentrations.The results show:In the April,choose the cuttings of secondary leaves have germinated about 2cm,the primary leaves do not fall off,in 180 cm high plastic greenhouses,the rooting rate is high,30.52%.But GGR concentration had no significant effect on rooting.For the different cuttings medium,the rooting rate of vermiculite was the best,28%.For different cuttings in May,the effect of GGR concentration on the cuttings was not significant,and the rooting rate of the cuttings that the tip of the primary coniferous cluster is not open to the bud was the highest,and the rooting rate was 87.33%...
