With beautiful streaks on surface of blades and the extreme resistance to the shade adaptation indoors,the coloured leaf of Aglaonema commutatun is a popular new species of potted flowers.Taken the rhizome with axillary buds of Aglaonema commutatum Schott 'Red Valentine'and 'Nightsparkle' as the explants,explore the effect of different disinfection time,different proportion of growth regulations and different medium type on bud initiation,callus induction,cluster buds,multiplication,rooting of buds and transplantation to establish effective isolated culture systems.Moreover,in order to preliminarily verify the physiological characteristics of leaf color change,study on the environmental factors affecting the leaf color of coloured leaf of Aglaonema commutatun was carried out to provide the scientific basis for the extensive introduction and cultivation and its application in garden of coloured leaf of Aglaonema commutatum.The main results of the experiment are as follows:1?Study on isolated culture of coloured leaf of Aglaonema commutatum(1)The optimal sterilization time for 'Red Valentine' and 'Nightsparkle' was 18 min and 15 min.The optimal callus induction medium was 1/2MS+0.5 mg/L TDZ+2.5 mg/L 2,4-D and 1/2MS+0.5 mg/L TDZ+2.0 mg/L 2,4-D,the callus induction rate was higher after optimizing medium.(2)The best cluster buds induction medium for 'Red Valentine' and 'Nightsparkle' was 1/2MS+0.5 mg/L TDZ,the bud induction efficiency was higher after optimizing medium and the optimal switching period is 30 days.(3)When the kinin is 6-BA,the optimal cluster buds multiplication medium for 'Red Valentine'and 'Nightsparkle' was MS+3.0 mg/L 6-BA+0.1 mg/L NAA and MS+4.0 mg/L 6-BA+0.1 mg/L NAA;When the kinin is KT,the optimal cluster buds multiplication medium was MS+4.0 mg/L KT+0.1 mg/L NAA and MS+5.0 mg/L KT+0.1 mg/L NAA.(4)The best rooting medium was 1/2MS+0.2 mg/L NAA.The best appropriate combination for naturalizing and transplanting was 2/3 turf+1/3 perlite.2.Study on the factors influencing the change of leaf color of Aglaonema commutatum(1)Under different light intensity,the change of leaf color was different of Aglaonema commutatum.With the increase of light intensity,the green color of leaf decreased,and the coloured plaques increased,the photosynthetic pigment decreased,while anthocyanin and soluble sugar increased.The photosynthetic pigment of leaf was the highest in the middle stage,while the anthocyanin was highest in the large stage.(2)At different temperature,the change of leaf color was also different.With the increase of temperature,the leaves became green and coloured plaques decreased,the content of photosynthetic pigment increased and the content of anthocyanin and soluble sugar decreased.The content of anthocyanin was higher under 15? than under 25? and 35?,the low temperature was beneficial to the synthesis of anthocyanin.(3)Under the proportion of light,the change of leaf color also showed a certain extent difference.When the proportion of red and blue light were 2:8,5:5 and 10:0,photosynthetic pigment content of Aglaonema commutatum were higher,and the content of anthocyanin and soluble sugar content were lower,the change of leaf color was not obvious.At 3:7 and 8:2 of red and blue light proportion,the photosynthetic pigment content of leaf of Aglaonema commutatum were lower,while anthocyanin and soluble sugar content were higher,the coloured spots of leaf increased.(4)By carring on correlation analysis between physiological indexes and environmental factors affecting leaf color change,the results showed that the contents of chlorophyll a and carotenoid of 'Red Valentine' were positively correlated with temperature,the content of anthocyanin and soluble sugar were extremely significantly or negatively correlated with temperature.The content of anthocyanin and soluble sugar of 'Nightsparkle' were very significantly or significantly positively correlated with the light intensity.There were significant positive correlations between chlorophyll a,chlorophyll a+b and temperature,significant or extremely significant negative correlation between anthocyanin and soluble sugar and temperature.
